| Objective: To investigate the Effect of EIF4G1 on the biological function and mechanism of lung squamous cell carcinoma.Methods: The relationship between EIF4G1 expression and lung squamous cell carcinoma was investigated by immunohistochemical technique;The effect of EIF4G1 on the biological function of lung squamous cell carcinoma was investigated by in vitro cell experiments.The effect of EIF4G1 on the growth of lung squamous cell carcinoma was verified by the tumor-bearing experiment in vivo,detection of signaling pathways by immunoblotting.Results:(1)The reslut about expression of EIF4G1 in Lung squamous cell carcinoma and the effect:(1)EIF4G1 expression in lung squamous cell carcinoma was higher than adjacent tissues(P<0.05),expression of EIF4G1 was different at stages and grads in patients tissues;(2)Within the 15.5-month survival period,the EIF4G1 low & medium expression group and the high-expression group had no significant difference in survival(P>0.05);After15.5 months of survival,the EIF4G1 low & medium expression group and the high-expression group had significant difference in survival time(P<0.05),and the high-expression group had lower survival than the lower & middle expression group;(3)EIF4G1 expression was negatively correlated with survival(r=-0.308,P<0.05),positively and lightly correlated with tumor volume of lung squamous cell carcinoma(r=0.195,P<0.05),no correlation with grads and stages of lung squamous cell carcinoma(P>0.05);(4)Stages of lung squamous cell carcinoma and tumor volume showed a positive weak correlation(r=0.319,P<0.05),and positive correlation with lymphoid positive number of lung squamous cell carcinoma(r=0.781,P<0.05);(5)Lung squamous cell carcinoma patiens showed positive status and weakly age correlation(r =0.175,P<0.05)。(2)The results of EIF4G1 expression on lung squamous cell carcinoma and its mechanism are as follows:(1)EIF4G1 expression in lung squamous cell carcinoma is higher than that in normal lung epithelial cells(P<0.05);(2)The expression of EIF4G1,cell viability,colony forming ability and invasion ability in the negative control group were significantly higher than those in the knockdown group(P<0.05);(3)Apoptosis rate of the cells in the EIF4G1 knockdown group was higher than negative control group,but there was no significant difference(P>0.05),the migration ability results also showed no difference(P>0.05);(4)EIF4G1 knockdown had cell cycle arrest in S phase,while knockdown group decreased Cyclin D1 expression;(5)Results in vivo showed EIF4G1 knockdown lung squamous cell carcinoma cells had smaller tumor formation volume and lower tumor weight than the vector group(P<0.05);(6)The group of EIF4G1 showed higher expression than the negative control group about AKT2/p-AKT.Conclusions:(1)EIF4G1 has an early role in predicting the prognosis of patients with lung squamous cell carcinoma;(2)The expression of EIF4G1 affects the biological function of lung squamous cell carcinoma cell,such as:proliferation,colony formation,invasion,cell cycle,apoptosis;(3)Knocdown of EIF4G1 is involved in PI3K/AKT pathway.EIF4G1 affects the development of lung squamous cell carcinoma. |
