Objective To study the protective effect and mechanism of berberine on myocardial infarction in old rats.Methods78 old rats(male,16 months old)were randomly divided into sham operation group,myocardial infarction saline injury group and myocardial infarction berberine treatment group.After opening the chest of all the old rats under general anesthesia,the sham operation group directly sutured the wound without damaging the heart,and the other two groups performed permanent ligation on the middle and upper 1 / 3 of the left anterior descending coronary artery.On the second day after operation,saline and berberine were injected intraperitoneally for 28 days,and the death and weight changes were recorded.On the 7th day,10 rats in each group were killed,and the level of inflammation was measured by cardiac tissue.The observation indexes included integrin α m(CD11b),interleukin-6(IL-6),chemokines 2(CCL2),tumor necrosis factor α(TNF-α).On the 28 th day,the remaining 10 rats in each group were assessed for cardiac function by echocardiography,and the heart tissues were stained with he and Masson after sectioning.Results(1)In the mortality and weight monitoring,compared with the myocardial infarction group,the mortality of berberine treatment group was significantly reduced,but there was no significant difference in weight.(2)In the echocardiographic monitoring experiment,Left ventricular ejection fraction(LVEF)was lower in the myocardial infarction group than in the sham group(P<0.001),the shortening rate of left ventricular short axis(LVFS)was also lower than that of the sham group(P<0.001).After berberine treatment,the heart function of the old rats after infarction was significantly restored.LVEF was significantly increased(P<0.001),and LVFS was significantly reduced(P<0.001).(3)HE staining showed that compared with the sham operation group,the myocardial infarction group had discontinuous tissue fracture.Berberine treatment can protect the structural integrity of cardiomyocytes.Masson staining showed that the degree of fibrosis in the myocardial infarction group was higher than that in the sham group(P<0.001).Berberine treatment can reduce the degree of fibrosis in myocardial tissue after myocardial infarction(P<0.005).(4)In terms of the level of tissue inflammation,the results showed that on the 7th day after berberine treatment,the inflammatory level of the injured heart was significantly lower than that of the myocardial infarction group.The results of flow cytometry showed that after berberine treatment,the number of CD11 b positive monocytes decreased(P <0.01);quantitative real-time polymerase chain reaction(Q-PCR)showed that the levels of IL-6 and TNF α genes were significantly reduced(P < 0.01),and the level of inflammatory chemokine CCL2 was also reduced(P < 0.05).(5)In terms of the molecular mechanism of berberine in the treatment of myocardial infarction,the results of Q-PCR showed that the level of PI3 K and Akt increased in Berberine group(P < 0.05).After treatment with PI3 K specific inhibitor(3-m A),the level of IL-6 and TNF α gene increased(P < 0.05).Conclusion Berberine treatment can promote the recovery of cardiac function in old rats after myocardial infarction,and its mechanism may activate PI3 K / Akt pathway to play an anti-inflammatory role,which lays a theoretical foundation for the clinical application of berberine in myocardial infarction rehabilitation. |
