Effect Of Acupuncture Combined With Yiqi Jianpi TCM Compound On Gastric Motility And RhoA-ROCK Signal Pathway In Diabetic Gastroparesis Rats

Objective To observe the effect of acupuncture combined with Yiqi Jianpi TCM compound on gastric emptying rate,small intestine propulsion rate,RhoA/ROCK signal pathway,p-MYPT1,P-MLC protein in diabetic gastroparesis（DGP）rats.To investigate the mechanism of this treatment on gastrointestinal motility in DGP rats,To provide a better method and basis for the treatment of DGP.Methods DGP model was prepared by the method of streptozocin（STZ）tail vein injection and irregular feeding with high sugar and fat for 8 weeks.The model rats were randomly divided into model group,acupuncture group,Yiqi Jianpi Chinese medicine compound group（TCM group）,acupuncture combined with Yiqi Jianpi Chinese medicine compound group（Acupuncture combined with TCM group）and domperidone group（Western medicine group）.the acupuncture group was given acupuncture30 minutes at Zusanli and Sanyinjiao daily.the TCM group was given Yiqi Jianpi Chinese medicine compound 5g/kg,Acupuncture combined with TCM group was first administered with Yiqi Jianpi traditional Chinese medicine compound 5g/kg,followed by acupuncture（operation is the same as the acupuncture group）;the western medicine group was grabbed and fixed for 30 minutes every day,and given daily domperidone3.15×10^-3g/kg;The blank group and model group were grabbed and fixed for 30 minutes every day,and given 10 ml / kg of normal saline to the stomach once a day,All treatment lasted for 4 weeks.Blood glucose,body weight,diet,hair color and mental state were recorded during the experiment.After the treatment,gastric emptying rate and small intestine propulsion rate were detected by phenol red method,pathological changes of gastric antrum mucosa were observed by HE staining,contents of Ras homologous genome members A（RhoA）and Rho protein-related curl spiral kinase-2（ROCK2）in gastric antrum tissue were measured by immunohistochemistry,and Western blotting was used to detect the expression of phosphorylated myosin phosphatase target subunit 1（p-MYPT1）and phosphorylated myosin light chain（p-MLC）protein in gastric antrum.Results Compared with the blank group: the symptom score and blood glucose of the model group were significantly higher（P<0.01）,weight loss was significant（P < 0.01）,gastric emptying rate,intestinal propulsion rat slowed down significantly（P<0.01）,HE staining showed that the mucosa level of gastric antrum was not obvious,the glands were relatively less,the arrangement was disordered and evacuation,and the blood vessels were reduced,and the expression level of RhoA,ROCK2,p-MYPT1 and p-MLC protein were significantly lower（P<0.01）.Compared with the model group:（1）Rats in each treatment group had reduced symptom scores and the body weight increased（P <0.01）,and the blood glucose in each group of Chinese medicine treatment was significantly reduced（P <0.01）,Although the blood glucose in the western medicine group was reduced,the difference was not statistically significant（P>0.05）.（2）The gastric emptying rate,small intestine advancing rate were increased in each treatment group（P<0.05 or P<0.01）,Among them,the gastric emptying rate of Acupuncture combined with TCM group was significantly higher than that of other treatment groups（P < 0.01）.（3）HE staining showed that the pathological changes of gastric antrum mucosa of rats in each treatment group were improved in different aspects,and the comprehensive improvement was most obvious in the Acupuncture combined with TCM group.（4）The expression levels of RhoA,ROCK2,p-MYPT1,and p-MLC protein in gastric antrum tissue of each treatment group increased（P <0.05 or P <0.01）,and p-MYPT1,p-MLC protein were significantly increased in the Acupuncture combined with TCM group（P<0.01）.Compared with other treatment groups,The differences were statistically significant（P<0.01 or P<0.01）.Conclusions（1）Acupuncture combined with Yiqi Jianpi TCM compound treatment can enhance gastrointestinal motility of DGP rats and improve related symptoms of DGP rats,and the effect is better than using Chinese medicine compound alone.（2）The mechanism of acupuncture combined with Yiqi Jianpi TCM compound to enhance gastrointestinal motility may be related to up-regulation the RhoA / ROCK signal pathway in gastric antrum,increase expression of p-MYPT1,p-MLC protein,and promote gastric smooth muscle contraction.
