| Objective:To investigate the inhibitory effect of mahuangdingtongtang(MTD)on vascular endothelial structure destruction and oxidative stress in atherosclerosis(AS)model rats,to clarify the regulatory effect of MTD on the risk factors,related vasoactive substances and metabolites of AS,and to elucidate the mechanism of MTD inhibiting the occurrence of AS and to provide experimental basis for clinical use of MTD in the treatment of AS.Methods:Forty-two male SD rats of clean grade were divided into 7 in the normal group and 35 in the AS model group.The normal group of rats was fed with normal diet for 10 weeks,and the AS model group of rats was intraperitoneally injected with vitamin D3 600,000 IU/kg at one time during modeling,and additionally injected with vitamin D3 100,000 IU/kg at the 4th and 8th weeks after the start of modeling,two days for injection,and fed with high fat diet for 10 weeks.After successful modeling,the rats in the AS model group were divided into five groups by random blind selection,which were 7 rats in low,middle and high dose groups of MTD,Atorvastatin group and model group.The normal group and the model group were intragastrically given the same dose of normal saline,the atorvastatin group was intragastrically administered with the corresponding dose of atorvastatin calcium tablets,and the MTD group was intragastrically administered with the corresponding dose of MTD,for 4 weeks.After 28days of treatment,histopathological changes in the rats aorta were examined by H&E staning;by biochemical method to detect the rat serum catalase(CAT),malondialdehyde(MDA),superoxide dismutase(SOD),glutathione peroxidase(gsh-px),and the blood lipid content;ELISA were used to observe the serum content of endothelin 1(ET-1),nitric oxide(NO),oxidized low density lipoprotein(ox-LDL).Results:1.The results of staining of rats aorta showed that the model group of rats,plaque formation was observed in the aorta,with varying numbers of membrane cells in the plaques,and the aortic intima was thickened,accompanied by endothelial cell injury and shedding,exposure of deep collagen fibers,and variable thickening of the internal elastic lamina.2.The results of staining of rats aorta showed that MTD could reduce atherosclerosis and vascular endothelial injury after model establishment in rats,and the atherosclerosis index(AI value)was significantly lower than that in the model group.3.Atorvastatin group and all group of MTD could improve the blood lipid index of rats,and the medium and high dose groups of MTD could reduce the serum TC and TG indexes(P<0.05),and the efficacy was equivalent to atorvastatin group.4.MTD could reduce the activity of MDA、ox-LDL,increase the content of SOD,CAT and GSH-PX in serum,and the effect of MTD in the low and medium dose groups was significant(P<0.05).5.MTD can increase the level of serum NO,decrease the release of serum ET-1,improve the vascular endothelial dysfunction and lipid adhesion,and has a better effect in anti-atherosclerosis.Among them,medium dose group of MTD had the most significant effect.Conclusion:MTD has a good anti-atherosclerotic effect,promote the regulation of lipid metabolism,and regulate the level of oxidative stress in rats.increasing the serum NO content,decreasing ET-1 content,be better restore NO system. |
