Objective Study the immunomodulatory effects of vancomycin and LYWW01,determine whether their effects is different and explore the mechanism of which,so as to provide certain guidance for the clinical use of vancomycin and establish the foundation for further study of LYWW01.Methods LPS(lipopolysaccharide)was used to stimulate RAW 264.7 macrophage to establish an in vitro sepsis model.Expression of cytokines such as TNF-α,IL-1β and IL-6 was detected by qPCR.Flow cytometry was used to detect the effect of vancomycin and LYWW01 on phagocytosis of RAW264.7 macrophage.Effects of vancomycin and LYWW01 on the release of reactive oxygen species was evaluated with DCFH-DA probe by flow cytometry.Western Blot was used to detect the LPS-induced synthesis of iNOS and activation of MAPKs,NF-κB and Akt.Results Vanomycin and LYWW01 had no effect on the synthesis of NO and the release of iNOS induced by LPS,and neither had an obvious effect on the expression of TNF--α.However,the high concentration of vancomycin inhibited the expression of IL-6,while the high concentration of LYWW01 had no significant effect on the the expression of IL-6,and the low concentration of LYWW01 promoted the expression of IL-6.Meanwhile,both of vancomycin and LYWW01 enhanced the phagocytosis of LPS-activated RAW264.7macrophage and inhibited the rise level of LPS-induced ROS.Conclusion Both vancomycin and LYWW01 have immunomodulatory effects,and their functions are relatively similar with slight differences.The mechanism of their immunomodulatory effects is related to their inhibition of p38 MAPK and Akt phosphorylation.
