This thesis consists of three chapters and the work during the master’s study is summarized.In the first chapter,the chemical constituents of Dobinea delavayi were dealt with.In the second chapter,the biological activities of D.delavayi were presented.The research progress of chemical constituents of Anacardiaceae was summarized in the last part.Objective:In this study,the chemical constituents of the roots of D.delavayi were studied to identify the monomeric constituents withβ-hematin formation inhibitory activity and to find the leading compounds or novel natural compounds with antimalarial activity.Methods:The compounds were isolated and purified by various column chromatography,and their structures were elucidated on the basis of spectral data and physicochemical properties.The biological activities of the extracts were measured byβ-hematin formation inhibition test.Results:Thirty-nine monomeric compounds were obtained from ethyl acetate fraction of the 80% ethyl alcohol extract the roots of D.delavayi.The compounds were identified as dodelate A-M(1-13),3β-angeloyloxy-4α-hydroxy-eudesm-7(11)-en-8β,12-olide(14),3β-angeloyloxy-4α-hydroxy-eudesm-7(11)-en-8α,12-olide(15),dobininⅠ(16),dobininⅡ(17),3β-angeloyloxy-4α-hydroxyl-eudesm-7(11),8(9)-dien-8,12-olide(18),3β-angeloyloxy-4α-acetoxy-eudesm-7(11)-en-8α,12-olide(19),3β-angeloyloxy-4α,8β-dihydroxy-eudesm-7(11)-en-8α,12-olide(20),3β-angeloyloxy-4α-acetoxy-8β-hydroxy-eudesm-7(11)-en-8α,12-olide(21),spathulenol(22),2,6,3’,4’-tetrahydroxy-4-methoxylbenzophenone(23),2,6,4’-trihydroxy-3’,4-dimethoxybenzop-henone(24),1,6,7-trihydroxy-3-methoxyxanthone(25),2,6,4’,6-trihydroxy-4-methoz-xybenzophenone(26),mangiferin(27),3,4,2’,4’-tetrahydroxydihydrochalcone(28),3,4,2’,4’,α-pentahydroxychalcone(29),aureusidin(30),butin(31),24-metbylenecycl-oartanol(32),6β-hydroxystigmast-4-en-3-one(33),glyceroylmono-palmitate(34),β-sitosterol(35),gallic acid(36),bisphenol A(37),methyl hexadecanoate(38),and daucosterol(39).The antimalarial activities of the D.delavayi roots crude extract and different solvent extraction fractions of crude extract were evaluated byβ-hematin formation inhibition assay,and only crude extract,n-butanol soluble fraction and water soluble fraction showed the different activity.The antimalarial activities of 19compounds separated from ethyl acetate soluble fraction were detected as well,and only 6 compounds showed inhibitory activity.The compounds 29~31 have significant activity with IC50 values of 61.5±3.4,56.0±3.4 and 120.3±4.0μg/mL.Conclusion:Compounds 1~13 and 23~25 from D.delavayi were identified as new compounds.Compounds 22,26,28~33,36 and 37 were isolated from the Dobinea for the first time.Terpenoids are the major constituents of the plant.The results of bioactivity test showed that the compounds with phenolic hydroxyl group had betterβ-hematin inhibitory activity,which indicated that the plant had certain potential in antimalarial activity.This study provides a scientific basis for the deeper study and application of D.delavayi in the future.
