Objective: To investigate the changes in the number of natural killer cells in peripheral blood and peritoneal fluid and the changes in cell functions such as secreted cytokines and cytotoxicity at different time points when mice develop sepsis.Methods: 48 male C57 BL / 6 mice were randomly divided with 6 mice in each group.The mice were subjected to cecal ligation and puncture as the sepsis model.Peripheral blood and abdominal cavity were taken at 0,6,24 and 48 hours after surgery Washing fluid was used to measure the proportion of natural killer cells by flow cytometry.Plasma and peritoneal lavage fluid were used as an enzyme-linked adsorption test to detect the levels of cytokines: interleukin 15 and granzyme B.Record the weight changes of mice at various time points,and take liver,spleen and lungs at each time point for staining pathological sections for observation and analysis.Results: After the establishment of the sepsis model,the change in body weight was statistically significant among the groups(P<0.05),indicating that the model was successfully established and the weight loss trend was significant.There was no significant difference in the number of lymphocytes between the two groups in the 0h group(P>0.05).There was a statistically significant difference between the two groups in the 6h,24 h and 48 h groups(P<0.05),indicating that the lymphocytes circulated from the periphery Has migrated to the local inflammation site.The proportion of NK cell flow cytometry in lymphocytes showed that there was a statistically significant difference between the two groups at 0h,6h,24 h and 48h(P<0.05).NK cells migrated from the peripheral circulation to the inflammatory response of the abdominal cavity Participate in acute immune response.In the 0h,6h,24 h and 48 h groups,the levels of IL-15 and granzyme B were statistically significant in the comparison of plasma and peritoneal lavage fluid(P<0.05),indicating that the cytokine concentration first increased during sepsis After high,it gradually decreases,which is consistent with the fluctuation trend of the number of NK cells.Conclusion: When CLP-induced sepsis and septic shock in mice,the number of mononuclear lymphocytes in the peripheral circulation decreases,while the number of inflammatory cells in the local inflammation site increases,and the NK cells in the peripheral blood decrease,and the NK cells in the abdominal lavage fluid increase NK cells are chemotactically induced to migrate from peripheral blood to the abdominal cavity to participate in the formation of inflammatory pathological processes of septic shock.The levels of IL-15 and granzyme B also change with the migration of NK cells,and the level of change first increases After the decrease,it plays an important role in maintaining the stability of NK cells and its killing function.
