Enzyme-linked immunosorbent assay(ELISA)is a rapid detection method that combines the antigen-antibody immune reaction with the enzyme catalytic reaction,and is also a common method used to detect disease biomarkers in the hospital.This method mainly uses 96 microplate for high-throughput biochemical reactions.The microplate reader is a professional instrument for analyzing the results of ELISA reactions in microplates,which is a spectra analysis instrument.Due to containing precise optical components and complex optical path,microplate reader is large,expensive,and complicated to operate,more importantly,the process of the data analysis depends on the computer.Therefore,it is difficult to apply to rapid on-site detection of disease biomarkers.With the widespread popularity of smartphones and the rapid improvement of its performance,the number of analysis and detection instruments based on smartphones has been increased year by year.These instruments have the advantages of light weight,portability,low price and easy operation,thus having great possibility was to be utilized in rapid on-site diagnosis of disease.In this paper,smartphone is used as an analysis and detection tool,and a portable microplate analyzer was designed in combination with 96 microplates.A large-aperture lens was firstly used in this analyzer to assist smartphone to achieve a uniform imaging of the whole 96 microplate.On this basis,the designed optical device and the custom-developed Android app are used to perform automatic colorimetric analysis.The developed analyzer has realized high-throughput rapid quantitative detection of some biomolecules,which can be used in the detection of disease biomarkers.The main research contents of this article are as follows:1.After studying the optical principle and specific optical path of the smartphone camera imaging,a large-aperture lens was designed to assist smartphone to achieve a uniform imaging of the whole 96 microplate.A microplate analyzer based on smartphone was developed,and it includes designed potable optical devices and custom-designed smartphone applications,which could quantitatively detect the test samples in microplate digitally and automatically.2.High-throughput rapid quantitative detection of human serum albumin(HSA)was realized through the developed analyzer.The detection results of HSA in patient serum samples are consistent with the detection results from the automatic biochemical analyzer in the hospital.The detection range was 21.45 ~60.06 g/L,which can totally meet the demands of disease detection in the hospital.3.The developed analyzer realized multiplex rapid detection of neonatal infectious disease biomarkers including C-reactive protein(CRP),serum amyloid(SAA)and procalcitonin(PCT).The detection range of the analyzer for these three indicators were 150 ~ 2400 μg/L,50 ~ 800 μg/L and 75 ~ 1200 ng/L,respectively.The actual serum samples achieved from local hospital were tested by using this analyzer,and the detection results showed great consistency with the results obtained by the microplate reader,implicating the developed analyzer could be used for the multiplex rapid detection of three biomarkers in the hospital.
