| Objective:In recent years,it has been proved that microRNAs can be detected stably in plasma /serum,and play an important role in the diagnosis of esophageal cancer,gastric cancer,pancreatic cancer,colorectal cancer,liver cancer and other digestive system tumors.The purpose of this study is to further explore the specific mechanism of microrna-1290(mirna-1290)targeting the expression of N-myc downstream regulated gene 1(NDRG1)and interleukin-6(IL-6)involved in the invasion and migration of colon cancer cells.Methods:We examined the expression of IL-6 and mirna-1290 in colon cancer and its adjacent tissues,colon cancer cells(HT-29,SW480 and SW620)and normal colon epithelial cells of hcoepic by immunohistochemistry and RT-PCR.We designed and constructed Mirna-1290 mimics and mirna-1290 inhibitors to enhance and inhibit the expression of mirna-1290 in colon cancer cells.RT-PCR was used to test the transfection effect.We used Western blot and ELISA to detect the protein changes of p-nf-κ B,NF-κ B,NDRG1,E-cadherin,vimentin and IL-6 exocrine in colon cancer cells.With the treatment of small interfering RNA-IL-6(siRNA-IL-6)or neutralizing antibody,the effect of miRNA-1290 in colorectal carcinoma cell migration was assessed by wound healing test,and cell invasion was tested by MatrigelTM invasion assay.Results:The expression levels of IL-6 and miRNA-1290 in colorectal carcinoma tissues and cells were significantly higher as compared with the adjacent normal tissues and HCoEpiC,and a remarkably positive correlation existed.Upregulation of miRNA-1290 by mimic inhibited the expression of NDRG1 and further induced the activation of NF-κB pathway and epithelial mesenchymal transformation(EMT),while Down regulation of miRNA-1290 restored the expression of NDRG1,E-cadherin and inhibited the formation of p-nf-κ B and vimentin.miRNA-1290 promotes the activation and secretion of NF-κ B signal and IL-6 by targeting down regulating the expression of NDRG1.Scratch test showed that up regulating the expression of miRNA-1290 could enhance the migration ability of colon cancer cells,while the cell healing rate was reversed after knocking down IL-6.miRNA-1290 mimic treatment promoted the invasion of colon cancer cells,while antibody neutralization of IL-6 could effectively reduce the number of invasive cellConclusion:miRNA-1290 can promote the proliferation,migration and invasion of colorectal cancer cells;miRNA-1290 in colon cancer cells promote NF-κ B / IL-6-induced migration and invasion by targeting NDRG1. |
