[Objective] To probe the effect of single nucleotide polymorphism of matrix metalloproteinase-3(MMP-3)gene and cerebral infarction and the stability of atherosclerotic plaque.[Methods] 546 patients with cerebral infarction in our Department were collected continuously.According to the results,they were divided into three groups: the unstable plaque group(338 cases),the stable plaque group(136 cases)and the non plaque group(72 cases).350 physical examinees in the physical examination center of our hospital were acquired continuously as the control group.we examined 4 genes of MMP-3,including rs602128、rs679620、rs520540、rs3025058),using matrix-assisted laser desorption/ionization time of flight mass spectrometry.23.0 statistical software and GMDR betav0.7 software were used for analysis.[result] 1.The male,hypertension,diabetes,age,systolic blood pressure,diastolic blood pressure,fasting blood glucose and homocysteine in the cerebral infarction group were all higher than those in the control group,while the cholesterol,high-density lipoprotein and low-density lipoprotein were all lower than those in the control group(P < 0.05).There was no significant difference in smoking,drinking,triglyceride and uric acid between the two groups(P > 0.05).2.rs602128 gene polymorphism was significantly different between cerebral infarction group and control group(P<0.05).There was no significant difference in rs679620、rs520540、rs3025058 between cerebral infarction group and control group(P>0.05).3.MMP-3rs602128,rs679620,rs520540 and rs3025058 gene polymorphisms had no significant difference between unstable and stable plaques(P>0.05).4.Logistic regression analysis showed that age,hypertension,diabetes and homocysteine were independent risk factors for cerebral infarction,while,men,hypertension and homocysteine were independent risk factors for unstable plaque of cerebral infarction.The mutation of rs602128 G site was negatively correlated with the disease.Compared with the AA type without mutation,the incidence of GG type cerebral infarction was reduced(OR:0.031,95% CI : 0.007-0.136).GMDR analysis showed that there was significant gene-gene interaction between rs602128 、 rs679620,and rs602128 、 rs679620 、rs520540 in cerebral infarction(the testing balanced accuracy was 61.80%,cross validation consistency was 10 / 10,P = 0.001),which could reduce the incidence of cerebral infarction(OR:0.698,95%CI:0.506-0.963;OR:0.087,95%CI:0.042-0.178).There was no significant gene gene interaction between rs3025058,rs679620,rs602128 and rs520540 in plaque group.[Conclusion] MMP-3 rs602128 gene polymorphism may be a protective factor for cerebral infarction,which can further reduce the incidence of cerebral infarction by interacting with rs679620、rs3025058. |
