Objective: To construct lnc RNA/mi RNA/m RNA regulatory networks related to macrophage polarization,The role of lnc RNA AK141955/mi R-124-3p/Sema6 d in the polarization of macrophages was investigated,To investigate the molecular mechanism of lnc RNA Ak141955 regulating mi R-124-3p/Sema6 d expression in macrophage polarization through the ce RNA mechanism,To elucidate the role of lnc RNA AK141955/mi R-124-3p/Sema6 d axis in regulating macrophage polarization in viral myocarditis.Methods: Bioinformatics software was used to construct the ce RNA regulatory network in the process of macrophage polarization,The key signal axis lnc RNA/mi RNA/m RNA was screened by q RT-PCR;Dual luciferase assay and RIP assay were used to verify the targeting relationship between mi RNA and lnc RNA and between mi RNA and m RNA;Rescue experiment was conducted to verify whether lnc RNA regulates mi RNA/m RNA expression through the ce RNA mechanism in vitro.Finally,the expression of lnc RNA was inhibited in mice to observe the role of lnc RNA in viral myocarditis,To clarify the role of lnc RNA AK141955/mi R-124-3p/Sema6 d axis in regulating macrophage polarization in viral myocarditis.Results: Bioinformatics software was used to successfully construct the ce RNA regulatory network that regulates the polarization of macrophages,lnc RNA AK141955/mi R-124-3p/Sema6 d,a key signal axis regulating the polarization of macrophages,was successfully screened by in vitro functional tests,Dual-luciferase and RIP experiments confirmed that mi R-124-3p had a targeted regulatory relationship with lnc RNA AK141955 and mi R-124-3p had a targeted regulatory relationship with Sema6 D,In vitro,lnc RNA AK141955 was demonstrated to regulate the expression of mi R-124-3p/Sema6 d through the ce RNA mechanism through the rescue experiment.Finally,by inhibiting the expression of lnc RNA AK141955 in mice,it was found that lnc RNAAK141955 could promote the progression of viral myocarditis.Conclusions: lncRNA AK141955 can regulate the expression of mi R-124-3p/Sema6 d through the ce RNA mechanism and play an important role in the process of macrophage polarization,lnc RNA AK141955/mi R124-3p/Sema6 d axis promotes viral myocarditis by regulating macrophage polarization. |
