Background:Prostate cancer(PCa)cells can communicate with other cells through exosomal miRNA,which plays a crucial role during the progression of prostate cancer.In recent years,more and more researchers have focused on the role of exosomal miRNAs in advanced prostate cancer.Some studies have shown that exosomal mir-375 is closely related to the occurrence and development of prostate cancer,but the specific molecular mechanism is still unclear.Methods:RNA-seq and qRT-PCR were used to screen the differential expression of serum exosomal miRNAs in prostate cancer patients with bone metastasis and localized prostate cancer patients.Then serum exosomal mirna-3 75 of prostate cancer patients was extracted,identified and expanded validation.The role of exosomal miRNA-375 in promoting the progression of prostate cancer was studied by collecting clinical data for statistical analysis,bioinformatics analysis,Western blotting,edu cell proliferation assay,Transwell assay,luciferase reporter assay,and its molecular mechanism was prelimially discussed.Results:miR-375 was significantly up-regulated in advanced PCa,and was closely related to serum PSA level,Gleason grade and PCa bone metastasis status.In addition,miR-375 secreted by tumor can promote the proliferation and invasion of prostate cancer cells.We also confirmed that miR-375 can promote the progression of PCa by targeting DIP2C to regulate Wnt signaling pathway.Conclusion:miR-375 may regulate Wnt signaling pathway by targeting DIP2C and promote the progress of PCa.
