With the improvement of living standards,obesity has become a global health problem.Western medical research believes that obesity is caused by excessive accumulation of body fat,its causes are closely related to dietary factors,social factors,metabolic factors,and gut microbiota factors.Obesity is also closely associated to diabetes,hypertension,hyperlipidemia,and other diseases,which can cause severe health problems.Clinically,the treatment of obesity by western medicine is mainly medical treatment,surgical treatment,meal replacement treatment,and other methods,which have achieved specific therapeutic effects.However,due to its side effects and contraindications,it cannot be promoted and applied to an extensive range of people.The earliest understanding of obesity in traditional Chinese medicine originated from the "Huang Di Nei Jing",which records that obesity is located in the spleen and stomach,and is related to the stop of the body’s water,dampness,and phlegm.Therefore,obesity often manifests as symptoms of spleen deficiency and wetness.The treatment of obesity in traditional Chinese medicine mainly includes Chinese herbal compound treatment,acupuncture treatment,tuina cupping,and other auxiliary treatment methods,which have good clinical effects,but suffer from a traditional Chinese medicine prescription that is widely applicable to most people.The treatment mechanism of obesity has not yet been fully elucidated.YiHeJianFeiCha used for our subject is an empirical formula used in the treatment of obesity by the folks.It has played an excellent effect on long-term clinical application.Its main components are:Coicis semen 30g,armeniacae semen amarum 10g,nelumbinis folium 10g,pogostemonis herba 10g,sojae semen praepamiceum 10g,cichorii herba cichorii radix 10g,pu’er tea 10g,poria 15g,the composition is in a patent application,patent number cn110585368a.Coicis semen is the prime medicine,with a peaceful nature,good hydration without damaging the true yin,and its function is to invigorate the spleen and dispel dampness;poria,cichorii herba cichorii radix,and nelumbinis folium are chen yao,help the coicis semen to absorb moisture,and at the same time,it has the effect of strengthening the spleen and removing fat.Armeniacae semen amarum,sojae semen praepamiceum,pogostemonis herba,and pu’er tea are all adjuvants,which can invigorate the stomach and eliminate food,promote water and neutralize,and have the effect of invigorating the stomach and removing food while eliminating dampness.Most of its prescription drugs are Chinese medicinal materials with the same medicine and food,without apparent cold and heat bias,and are suitable for popularization and application among the population.To confirm the effect of YiHeJianFeiCha on preventing and curing obesity,our research group conducted a study on the lipid-lowering effect of YiHeJianFeiCha for weight loss through preventive and therapeutic administration experiments.The specific methods and results are as follows:In the preventive dosing experiment,30 SPF mice aged 4-6 weeks were selected,ten mice in each group,and they were divided into a normal diet group,a high-fat diet group,and a high-fat diet administration group.According to the weight of the mouse,the intragastric administration was administered once a day.The weight of the mouse was recorded weekly.The experimental results showed that at the end of the experiment,the body weight of the mice in the normal diet group was 27±1.6g,the body weight of the mice in the high-fat diet group was 35.2±4g,and the body weight of the mice in the high-fat diet group was 26.7±1.3g.Compared with the body weight of mice in the high-fat diet group,the body weight of the mice in the high-fat diet group was significantly different(P=0.00 03,P<0.01),and there was a statistical difference.Moreover,the difference between the normal diet group and the high-fat diet administration group was not significant(P=0.69,P>0.05).Conclusion:YiHeJianFeiCha can prevent the occurrence and development of obesity.In the therapeutic administration experiment,40 SPF mice aged 4-6 weeks were selected,ten mice in each group,they were divided into normal diet group,normal diet administration group,high-fat diet group,and high-fat diet administration group.The mice were given eight weeks of non-intervention feeding,and then the high-fat mice were given intragastric administration once a day.The weight of the mice was recorded every week.The experimental results show that at the end of the experiment,the normal diet group:28.4±2.1g,the normal diet group:27.4±1.3g,the high-fat diet group:47.6±2.7g,the high-fat diet group:36.3±2.5g.Except that the difference between the normal diet group and the normal diet administration group was not significant(P=0.6252,P>0.05),the differences between the other groups were very significant(P<0.01).Conclusion:YiHeJianFeiCha can quickly and effectively treat the occurrence and development of obesity.To study the lipid-lowering mechanism of YiHeJianFeiCha for weight loss,we conducted related studies on mice in the preventive administration experiment and the therapeutic administration experiment.The specific methods and results are as follows:the body fat rate of the two experimental mice was analyzed by the nuclear magnetic resonance mouse body composition analyzer.The respiratory entropy of the two experimental mice was detected by the TSE mouse metabolic cage,and the fecal DNA was extracted.The kit performs cluster analysis of the gut microbiota of the intestinal feces of the two experimental mice and uses the blood biochemical analyzer to detect the serum samples of the two experimental mice,and the liver and adipose tissue of the two experimental mice,The histopathological staining analysis was carried out,the adipose tissue of the two experimental mice was screened for genes related to thermogenesis,and the Chinese medicine network pharmacology was used to analyze the YiHeJianFeiCha for weight loss.The experimental results showed that the body fat rate of mice treated with YiHeJianFeiCha for weight loss was significantly lower than that of mice in the high-fat diet group(P<0.01);The number and proportion of Firmicutes and Proteus in the intestines of mice in the high-fat diet group increased significantly,which was significantly different from that of the normal diet group(P<0.01).The abundance and proportion of Firmicutes and Proteobacteria in the high-fat diet group were different from those in the high-fat administration group Significant(P=0.002,P<0.01).This shows that the structure of the gut microbiota of the mice treated with YiHeJianFeiCha has been significantly improved compared with the mice in the high-fat diet group;The blood biochemical indicators(low-density lipoprotein LDL,alanine aminotransferase ALT,triglyceride TG,etc.)of the mice after treatment with YiHeJianFeiCha were significantly lower than those of the mice in the high-fat diet group,and the difference was statistically significant(P<0.01);the liver and fat pathological results of the mice treated with YiHeJianFeiCha for weight loss were significantly improved compared with the mice in the high-fat diet group.The results of the pyrogen gene screening showed that compared with the mice in the normal diet group,the expression level of PGC1-α mRNA in the fat cells of the mice on the high-fat diet group was significantly reduced,while the high-fat mice treated with YiHeJianFeiCha had a significant decrease in the expression level of PGC1-αmRNA.The mRNA expression level of PGC1-α in the high-fat mice treated with YiHeJianFeiCha group was significantly increased compared with the high-fat diet group,indicating that YiHeJianFeiCha treatment specifically enhances the mRNA expression level of PGC1-α.Conclusion:1.YiHeJianFeiCha changed the abundance and structure of the gut microbiota in mice,so improving the abundance of gut microbiota may be one of the targets of YiHeJianFeiCha for treating obesity;2.YiHeJianFeiCha for weight loss Tea treatment can activate the expression of PGC1-α,so activating the expression of PGC1-α and regulating lipid metabolism is one of the targets of YiHeJianFeiCha for obesity.This study conducted in-depth research on YiHeJianFeiCha for weight loss,and explored the mechanism and role of traditional Chinese medicine in the prevention and treatment of obesity.The study confirmed that YiHeJianFeiCha for weight loss through regulating the abundance of gut microbiota and activating PGC1-α expression regulates lipid metabolism,thereby preventing obesity.This study clarified the possible targets of YiHeJianFeiCha for reducing fat and losing weight,providing a new idea for the clinical treatment of obesity,and providing a scientific basis for the further development and research of YiHeJianFeiCha for weight loss.
