Effect Of Qingfeixiegan Decoction On Inflammation And Oxidative Stress In Type 2 Diabetic Rats

Abstract/Summary:
Objective: To explor studied the effects of Qingfei Xiegan Decoction(QFXG)on alleviating inflammation and oxidative stress in type 2 diabetes(T2DM)rats,and its possible mechanism of action.It aims to provide new ideas for QFXG in the treatment of T2 DM,so as to better play and tap the resource advantages of T2 DM in T2 DM.Methods: In this study,the traditional Chinese medicine QFXG was selected,and the medicinal solution was obtained by soaking,cooking,filtering,and rotary evaporation.High-fat and high-sugar feeding for 6 weeks,intraperitoneal injection of STZ after feeding to establish a T2 DM rat model with a dose of 30 mg/kg·BW.After 72 hours,the fasting blood glucose(FBG)level is higher than 16.7mmol/L as a successful model 。Successfully modeled rats were randomly divided into four groups,each with10 rats,including model group,high-dose QFXG group(1000 mg/kg),low-dose QFXG group(500mg/kg),positive control group(150 mg/kg).Continuous gavage for 8 weeks.During the test,the body weight and FBG of all rats were recorded every two weeks.On the last day of gavage,glucose tolerance level(OGTT),fasting insulin content(FINS),and insulin resistance were measured.Index(HOMA-IR),detected total cholesterol(TC),triglycerides(TG),high density lipoprotein(HDL-C),low density lipoprotein(LDL-C),and detected SOD,GSH,CAT,MDA related indicators,ELISA method was used to detect TNF-α,IFN-γ,IL-6,IL-1βand other inflammatory factors,Western Blot method to detect Myd88,p-p65,p-IκB-α and Expression of TLR4 protein.Results: Compared with the blank control group,the body weight of T2 DM rats treated with QFXG can be well controlled,and the levels of FBG,FINS,and HOMA-IR in T2 DM rats can be significantly reduced(P<0.05)and increased significantly OGTT of T2 DM rats.After QFXG treatment,it can significantly increase the level of TC,TG,LDL-C(P<0.05),and significantly increase the level of HDL-C.With QFXG high-dose group treatment,SOD,GSH,CAT activities in the liver tissue of T2 DM rats were significantly increased(P<0.05),and the content of MDA was extremely significantly reduced(P>0.05).After QFXG treatment,the contents of inflammatory factors TNF-α,IFN-γ,IL-6,IL-1β in the liver tissue were extremely significantly reduced(P<0.05),and the inflammatory pathway proteins Myd88,p-p65,and p-p65 in the liver tissue were also extremely significantly reduced.The expression of p-IκB-α and TLR4(P<0.05).Conclusion: QFXG can treat T2 DM rats,reduce fasting blood glucose and fasting insulin index,and improve oxidative stress and inflammation.The protective effect of QFXG on T2 DM rats can be achieved by inhibiting inflammation and oxidative stress.The mechanism may be involved Regulate the TLR4/Myd88/NF-κB signaling pathway to reduce the inflammatory response in T2 DM rats and play a therapeutic effect on T2 DM.
