| Objective: This study mainly explored the correlation between the poly morphism of Zinc finger protein 304(ZNF304)gene and TCM syndromes and clinical indexes of Ischemic Stroke.Methods: 774 cases ischemic stroke patients and 793 control samples were included.According to the "Diagnostic Criteria for Stroke Diagnosis(Trial)",the patients with ischemic stroke were differentiated.Massarray technology was used to genotype the ZNF304 gene polymorphism loci rs7250521 and rs35686482.The French St Ag O Capact blood coagulation meter and the HITACHI7600 automatic biochemical analyzer were used to detect the serum coagulation function and blood lipid levels of patients with ischemic stroke in accordance with standardized experimental procedures.Use PLINK software for genetic association analysis,and use SPSS software for other correlation analysis.Results:1.Hardy-Weinberg Equilibrium(HWE)test:The genotype frequencies of rs7250521(PHWE=0.192)and rs35686482(PHWE=0.625)in the control group conformed to the HWE.2.Genotype frequency distribution:Between the IS fire syndrome group and the control group,the difference in the genotype frequency of the ZNF304 gene rs7250521 locus was statistically significant(P=0.011);There were no statistically significant differences in the rs35686482 polymorphic locus of ZNF304 genege between wind syndrome,fire syndrome,phlegm syndrome,qi deficiency syndrome,blood stasis syndrome and the control group(P>0.05).3.Association study of ZNF304 gene polymorphism and IS and Its TCM syndromes:ZNF304 gene rs7250521 polymorphism is significantly associated with the risk of IS fire syndrome(additive model: Padj=0.003;dominant model: Padj=0.004).There was no statistically significant association between the rs35686482 polymorphism of ZNF304 gene and the risk of each syndrome of ischemic stroke(all P>0.05).ZNF304 gene rs7250521 and rs35686482 polymorphisms were not related to the risk of IS(both P>0.05).4.Correlation analysis between ZNF304 gene polymorphism and TCM syndrome score of IS patients:The ZNF304 gene rs7250521 polymorphism is significantly correlated with the IS patients’ fire syndrome score(additive model: Padj=0.020;dominant model: Padj=0.014].The ZNF304 gene rs35686482 polymorphism is no correlated with the IS patients’ syndrome scores(all P>0.05).5.Correlation analysis between ZNF304 gene polymorphism and clinical test indexes of IS patients:(1)The rs35686482 polymorphism of ZNF304 gene is significantly correlated with the DBP level of IS patients(additive model: Padj=0.010;dominant model: Padj=0.017);the rs35686482 polymorphism is significantly correlated with the FIB level(additive model: Padj=0.021;recessive model: Padj=0.037)of IS patients;rs35686482 polymorphism is significantly correlated with APO-B level(recessive model: Padj=0.042),LDL-C level(recessive model: Padj=0.041)of IS patients.(2)The rs7250521 and rs35686482 polymorphisms of ZNF304 gene were not statistically correlated with the levels of FPG and P2hPG in IS patients(all P>0.05);(3)The rs7250521 and rs35686482 polymorphisms of ZNF304 gene had nostatistically significant correlation with CRP,Hcy and UA in patients with IS(all P>0.05).Conclusions:(1)The rs7250521 polymorphism of ZNF304 gene may affect the occurrence on the fire syndrome of ischemic stroke.(2)The rs35686482 polymorphism of ZNF304 gene may affect the blood pressure level,coagulation function and blood lipid metabolism in patients with IS. |
