| Objective: Observe the movement of acupuncture in the cognitive function of Va D rats,the hippocampus PI3K-AKT signaling pathways downstream of key protein expression and apoptosis protein Caspase-3,the influence of the expression level in serum in the animal experiment on the basis of further explore movement acupuncture with conventional western medicine therapy in patients with Va D clinical efficacy,safety performance and its impact on Caspase-3 protein levels in serum.So as to understand the mechanism of action and curative effect of sports acupuncture in treating Va D,and provide objective and effective basis for running acupuncture in treating Va D.Methods:1.Animal experiment: the experiment to healthy male SD rats as the research object,Olsson method is adopted to establish the animal model of Va D rats model of postoperative survival is to be built by water maze test to test whether building success,behavioral and building successful Va D rats were randomly divided into 4 groups,model group,the first needle clumps,sports training and sports acupuncture group,each group of eight only.Then,8rats separated without ligation of the common carotid artery were randomly selected as the sham operation group and 8 healthy male SD rats as the normal group.packet can begin after the completion of treatment,the head be intradermal needle acupuncture needle clumps serum protein expression.2.Clinical experimental studies:Selection into the standard of 60 patients with Va D as experiment object,randomly divided into 2 groups,treatment group and control group,respectively,30 cases in each group,control group patients with Va D gives the basis of conventional western medicine treatment such as donepezil HCL,and was on the basis of the control group and treatment group with acupuncture treatment of movement,movement is acupuncture needle four god astute two holes around the will and shall exercise rehabilitation training at the same time,4 weeks of treatment cycles,respectively before and after treatment in patients with Va D related dementia rating scale(MMSE 、 HDS 、 ADL)and blood specimen collection,Serum caspase-3 protein expression levels in patients with Va D before and after treatment were detected by ELISA,and the safety performance of patients with Va D was evaluated during treatment.Results:1.Behavioral monitoring results: Compared with normal and sham operation groups,the escape latency of model group rats was significantly longer(P<0.05).Compared with the model group,the escape latency time of the exercise acupuncture group,the head acupuncture group and the exercise training group were shortened to different degrees(P<0.05),and the intervention effect of the exercise acupuncture group was the most obvious;In the space exploration experiment,compared with the normal group and the sham group,the first arrival time to the escape platform and the target quadrant residence time of the model group were significantly longer(P<0.05).Compared with model group,the first arrival time to escape platform and the target quadrant residence time of rats in exercise acupuncture group,acupuncture group and exercise training group were shortened to different degrees and prolonged to different degrees(P<0.05),and the intervention effect of exercise acupuncture group was the most obvious.2.Western-blot and ELISA results showed that compared with normal group and sham group,PI3 K and Akt proteins in hippocampal tissues of model group were significantly decreased,while serum Caspase-3 levels were significantly increased(P<0.05).Compared with model group,the protein levels of PI3 K and Akt in hippocampal tissues of rats in exercise acupuncture group,acupuncture cluster group and exercise training group were increased to varying degrees,and the level of serum Caspase-3 was decreased to varying degrees(P<0.05).The intervention effect was most obvious in the exercise acupuncture group.3.The results of clinical trials showed that there was no significant difference in serum caspase-3 levels between the control group and the treatment group before treatment(P>0.05).After exercise acupuncture combined with conventional Western medicine treatment,the expression of caspase-3 in serum of Va D patients was lower than that of the control group(P<0.05).The level of serum caspase-3 in Va D patients in 2 groups was lower than that before treatment(P<0.05).Moreover,the learning and memory ability and independent living ability of Va D patients improved to varying degrees after treatment compared with before treatment,especially in the treatment group.Conclusions:1.Animal and clinical experimental studies have found that the level of caspase-3 in serum of Va D patients and rats is increased,indicating that the abnormal expression of caspase-3 has a certain correlation with the occurrence and development of vascular dementia.2.The mechanism of exercise acupuncture in preventing and treating Va D may be through activating PI3K-AKT cell signaling pathway and down-regulating the expression of Caspase-3 protein. |
