| High-quality artificial skin has a wide range of applications in biotechnology and biomedical research,and the real-time,non-destructive quality inspection of artificial skin is of great significance.It is one of the key problems that need to be solved in the mass preparation of artificial skin.Optical coherence tomography(OCT)technology can perform real-time,non-invasive three-dimensional imaging inside biological tissues,which is very suitable for quality inspection of artificial skin.This study uses OCT technology to quantitatively study the parameters of the two dimensions of artificial skin’s morphological dimension and biological activity.The main research contents and innovations are as follows:1.We propose a method for quantitative analysis of multi-dimensional parameters of artificial skin based on OCT signal intensity characteristics.The artificial skin interface-point adaptive extraction algorithm is used to detect the thickness and roughness parameters in the morphological dimension of the tissue;the depth resolution algorithm of the single scattering model is used to calculate the OCT attenuation coefficient that characterizes the biological activity dimension.2.The accuracy of the thickness,roughness and attenuation coefficient measurement methods are verified through experiments.By comparing the results calculated by the algorithm with the results of H&E stained sections,the accuracy of the thickness calculated by the adaptive extraction algorithm for artificial skin interface points is verified;By comparing the measurement results of the algorithm in this paper with the image-based algorithm on the roughness standard model,the accuracy of the roughness value quantification of the algorithm in this paper is verified;By comparing the attenuation coefficient calculated by the artificial skin based on the single scattering model with the results of the MTT experiment,it is verified that the two have a high positive correlation.3.The OCT system was used to detect the artificial skin during the air-liquid culture cycle,and the change trend of the multi-dimensional parameters during the culture cycle was analyzed.The thickness distribution map and rough surface contour map of the artificial skin can clearly illustrate the culture state of the artificial skin tissue morphology,and the contour map of the attenuation coefficient value can show the distribution state of the artificial skin cell activity.4.Statistical analysis of the parameter changes trends of three different culture batches of artificial skin.Quantitative analysis of multi-dimensional parameters shows that the thickness and attenuation coefficient of the artificial skin is increasing during the air-liquid culture process;the surface roughness is in accordance with the skin keratinization,showing a trend of first decreasing and then increasing;Monitoring and feedback of the organizational morphology during the cultivation process,providing guidance and feedback for cultivating artificial skin.The method based on the OCT intensity signal can accurately quantify the change trend of the multi-dimensional parameters in the artificial skin culture process,providing an accurate detection method for the preparation of artificial skin. |
