The Study On The Rule Of Syndrome And Treatment And The Mechanism Of The Data Mining Prescription Of The Prestigious TCM Specialists In The Treatment Of Liver Cirrhosis

ObjectiveThrough collecting the prestigious TCM specialists treating liver cirrhosis medical records,using the traditional Chinese medicine inheritance auxiliary system to explore the prestigious TCM specialists’ treatment law and differentiation of treating liver cirrhosis,including the traditional Chinese medicine syndrome of liver cirrhosis,symptoms,prescription medication law and so on.At the same time,the network pharmacology research method was used to predict the mechanism of the data mining prescription in the treatment of liver cirrhosis,in order to provide a reference for the rational use of drugs and the development of new drugs for the treatment of liver cirrhosis.MethodsThrough the collection and collation of the prestigious TCM specialists related works on the treatment of liver cirrhosis cases,Input the collected patient information into the"traditional Chinese Medicine inheritance Assistance platform" system,This system is used to analyze and summarize the rules and the prescriptions of prestigious TCM specialists in the treatment of liver cirrhosis.Chemical constituents and related targets in the prescriptions obtained by TCMSP database retrieval,Search the Cenecards database for disease-related targets of cirrhosis,Combined with Venny2.1 analysis tools,Find the intersection of drugs and disease,And the prescription,liver cirrhosis,intersection target,drug active chemical composition data into Cytoscape3.7.1 software To construct a molecular interaction network of "liver cirrhosis disease-prescription chemical composition-intersection target" interaction,Cross-cutting targets are introduced into STRING database to establish protein interaction networks,Then the DAVID database was used for GO enrichment analysis and KEGG enrichment analysis.Outcome1.In the medical cases of liver cirrhosis treated by prestigious TCM specialists in China,there are more male patients,the proportion of men and women is nearly 5:1,and the incidence age of cirrhosis is the largest in the age stage of 41-50 years old,accounting for 35%of the cases.2.According to the frequency of drug use,the top 10 drugs were:Atractylodes macrocephala,Poria cocos,Salvia miltiorrhiza,Astragalus membranaceus,Biejia,Angelica sinensis,large abdominal skin,Bupleurum,Codonopsis,alisma;the proportion of Chinese medicine from high to low is:cold,warm,flat,cool,hot;the proportion of Chinese medicine from high to low is:sweet,bitter,symplectic,salty,sour,astringent;the proportion of return to the meridian is:spleen,liver,stomach,lung,kidney,heart,large intestine,gallbladder,bladder,small intestine,pericardium,Sanjiao.3.Using the auxiliary system of traditional Chinese medicine inheritance to analyze the data,it is concluded that among the 226 patients included in this study,the number of patients with Qi stagnation and blood stasis syndrome and liver stagnation and spleen deficiency syndrome is the largest,followed by damp-heat accumulation syndrome and spleen and kidney yang deficiency syndrome.4.Through data mining,the prescriptions of Chinese medicine for liver cirrhosis are Atractylodes macrocephala,Codonopsis,Poria cocos,Salvia miltiorrhiza,Biejia,Astragalus,Alisma,large abdominal skin,Radix Paeoniae Alba,Tujin,Angelica sinensis,Polyporus,etc.5.According to the data mining,8 potential new prescriptions based on the potential drug combination and core drug use rule of the famous old Chinese medicine in China are as follows:No.1 is composed of Codonopsis pilosula,Atractylodes macrocephala,Peony Peel,Astragalus membranaceus,Angelica sinensis and Fructus Aurantii;No.2 is composed of Forsythia Forsythia suspensa,Ma Lanju,Yanhuso,Xu Changqing;No.3 is composed of Fructus Aurantii,Fructus Fructus Aurantii,Salvia miltiorrhiza,aconite and Biejia.6.The effective components of the new prescription for liver cirrhosis were quercetin,luteolin,kaempferol,naringenin,mandshurica,isorhamnetin,7-methylisobuccinol,7methoxy-2-methylisoflavone,β-glutenin and so on.The prescriptions act on cirrhosis diseases in the form of multi-component,multi-target and multi-pathway,among which closely related targets are AKT1、JUN、RELA、MAPK3、MAPK1、IL6、MAPK14、MAPK8、VEGFA、EGFR、FOS.After GO enrichment analysis,it was found that the prescription was related to oxidative stress,inflammatory reaction,angiogenesis,cell metabolism,proliferation and apoptosis process through the regulation of toxic substances,inorganic substances,lipopolysaccharide.Through KEGG enrichment analysis,it was found that cancer signaling pathway,hepatitis B signaling pathway,PAKT signaling pathway,hepatitis C signaling pathway,NF-kB signaling pathway,VEGF signaling pathway and MAPK signaling pathway may be the main pathways in which prescriptions play a role.ConclusionThrough data mining,it is found that the prestigious TCM specialists for the treatment of liver cirrhosis in China is traditional Chinese medicine such as invigorating spleen and dampness,soothing liver and regulating qi,promoting blood circulation and removing blood stasis.The core drugs are Atractylodes macrocephala,Codonopsis,Poria cocos,Salvia miltiorrhiza,Biejia,Astragalus membranaceus,Alisma,large abdominal skin,Radix Paeoniae Alba,Tulip,Angelica sinensis,Polyporus,etc.Based on the research method of network pharmacology,the prescription acts on cirrhosis through multi-component,multi-target and multi-pathway,but the results of this study still need to be confirrmed by further experimental research.
