Objective:To study the effect and molecular mechanism of parecoxib on the migration,proliferation and invasion of SGC7901 gastric cancer cell line,and provide new ideas for the treatment of gastric cancer.Methods:(1)The half maximal inhibitory concentrations(IC50)of parecoxib on SGC7901 gastric cancer cell line was measured by Cell Counting Kit-8(CCK-8)and the appropriate drug concentrations gradient was selected.(2)MTT assay was used to detect the effect of different drug concentrations on proliferation of SGC7901 gastric cancer cell line for 24h and 48h;(3)Flow cytometry was used to detect the effects of different drug concentrations on the cell cycle of SGC7901 gastric cancer cell line for 24h and 48h;(4)The effect of different drug concentrations on the migration ability of SGC7901 gastric cancer cell line for 24h was detected by Transwell migration experiments and scratch healing;(5)The effect of different drug concentrations on the invasion ability of SGC7901 gastric cancer cell line for 24h was detected by Transwell invasion experiments;(7)The expression of NF-κB Rel A(p65)protein and its downstream proteins Bcl-2,ICAM1,MMP9,CYR61,E-cadherin and Cyclin D1 were detected by Western blot experiments.Results:(1)The CCK-8 results showed that IC50 of parecoxib on SGC7901 gastric cancer cell line were 201.1 umol/l for 24h and 141.8umol/l for 48h(P<0.05);(2)MTT assay showed that parecoxib could inhibit the proliferation of SGC7901 gastric cancer cell line in a dose-dependent manner(P<0.05);(3)Flow cytometry results showed that parecoxib could induce SGC7901 G0/G1 phase cell cycle arrest(P<0.05);(4)The results of Transwell migration experiments and scratch healing showed that parecoxib could inhibit the migration of SGC7901 gastric cancer cell line(P<0.05);(5)Transwell invasion experiments showed that parecoxib could inhibit the invasion of SGC7901 gastric cancer cell line(P<0.05);(6)The western blot results showed that NF-κB Rel A(p65)and its downstream proteins Bcl-2,ICAM1,MMP9,Cyr61 and Cyclin D1 were significantly decreased after parecoxib treatment in SGC7901 cells,meanwhile,E-cadherin expression was down-regulated(P<0.05).Conclusion:Parecoxib can inhibit the migration,proliferation and invasion of SGC7901 gastric cancer cell line by down regulating NF-κB signaling pathway.
