| Tulipa edulis(Miq.)Baker is a plant of the genus Tulip,which is mainly distributed in the Anhui,Henan,Jiangsu,Shandong and other provinces in China.It is used to treat swelling and pain of throat,scrofula,carbuncle,sore swelling,postpartum stasis,and treat goiter,whooping cough,esophageal cancer,gastric cancer in clinically.In this paper,the chemical composition and biological activity of T.edulis were systematically studied.Firstly,the active constituents of the T.edulis were separated and purified by silica gel column chromatography,MCI column chromatography,ODS column chromatography,sephadex column chromatography,reversed phase column chromatography,and high performance liquid chromatography.Secondly,the structures of compounds were confirmed by modern spectral techniques,such as ESI-MS,1H-NMR,13C-NMR,1H-1HCOSY,HSQC,and HMBC.Finally,the activity of the extracts and compounds were evaluated by the MTT method,anti-Aβaggregation and the protection of Aβinjured nerve cells experiments.A total of thirty-two compounds were isolated from the T.edulis,of which twenty-five compounds were identified as 2-O-diferuloylglycerol(1),1,3-O-diferuloylglycerol(2),tuli-palisol B(3),1-O-feruloylglycerol(4),1-O-p-coumaroy-2-O-feruloylglycerol(5),1,3-O-di-p-coumaroylglycerol(6),syringylglycerol(7),1-O-feruloyl-3-O-p-coumaroylglycerol(8),tuli-palisol C(9),2-O-p-coumaroylglyceride(10),1-O-p-coumaroylglycerol(11),dodecyl ferulate(12),tulipalisol A(13),nicotinamide(14),β-adenosine(15),uridine(16),1’,2’-dihydroxyethyl-methylpyrazine(17),uracil(18),adenosine(19),2-methyl-6-(2’,3’,4’-trihydroxy-butyl)pyrazine(20),3’-O-methyladenosine(21),colchicine(22),2-hydroxymethyl-3,5-dimethylpyrazine(23),2-hydroxymethyl-6-methylpyrazine(24),3,6-dimethylpyrazin-2-ol(25).Compounds 3,9 and13 are new compounds.Compounds 1-21,23-25 were isolated from T.edulis for the first time.The ethanol extract of T.edulis showed strong cytotoxicity against MCF-7 cells with IC50value of 16.87μg/mL.The chloroform extract showed strong cytotoxicity against MCF-7 and Hep G-2 cells with IC50 values of 16.16μg/mL and 9.64μg/mL,respectively.The ethyl acetate extract exhibited very weak cytotoxicity against to MCF-7,Hep G-2,and A549 cells(IC50values>30μg/mL).The Z2 showed potent cytotoxicity against MCF-7 cells with IC50 value of 20.67μg/mL.Compounds 1-14,17,20,21,23-25 exhibited very weak cytotoxic activity to MCF-7,Hep G-2,and A549 cells.Compounds 15,16,and 19 possessed strong cytotoxic activity against A549 cells with IC50 values of 28.08μg/mL,26.11μg/mL,27.85μg/mL,respectively.Compounds 16 and 19 showed strong cytotoxic activity against MCF-7 cells,with IC50 values of 13.51μg/mL and 14.81μg/mL,respectively.Compound 18 exhibited strong cytotoxic activity against Hep G-2 cells with IC50 value of 23.26μg/mL.Compounds 1,2-O-diferuloylglycerol(1)and 1,3-O-diferuloylglycerol(2)indicated the potent anti-Aβaggregation activity and protective effects on Aβ-damaged nerve cells.The research enriched the chemical library of plant resources of the T.edulis,which would provid a potential theoretical basis for the development and further utilization of the traditional Chinese medicine. |
