Objective:In this study,a new circular RNA circ PTBP3 was discovered through the GEO database.Then,we further analyze the expression,function and molecular mechanism of circ PTBP3 in colorectal cancer cells.This study aims to explore the role and molecular mechanism of circ PTBP3 in the occurrence and development of colorectal cancer.To provide experimental basis for the clinical development and application of circ PTBP3 as a molecular marker and therapeutic target for early diagnosis of colorectal cancer.Methods:The expression of circ PTBP3 in colorectal cancer cell lines and colorectal cancer tissues was detected by quantitative real-time PCR(q RTPCR)amplification.We screened out circ PTBP3(has_circ_0008192)with better verification effect.Meanwhile,in situ hybridization was used to verify the expression of circ PTBP3 in colorectal cancer tissues,and to analyze the relationship between circ PTBP3 and clinical characteristics.The knockdown model was constructed in colorectal cancer cell lines,and the expression of circ PTBP3 was knocked down by si RNA interfering with circ PTBP3 in colorectal cancer cell lines.CCK-8,plate clone formation,scratch healing and Transwell assay were used to observe the effects of circ PTBP3 on the proliferation,migration and invasion of colorectal cancer cell lines.q RT-PCR and Western blotting were used to detect the effects of circ PTBP3 knockdown on m RNA and protein levels of EGFR and MAP3K1.Results:We analyzed the GEO database GSE126094 and found that circ PTBP3 was highly expressed in colorectal cancer tissues.In situ hybridization experiments also showed that circ PTBP3 is highly expressed in colorectal cancer tissues,and the high expression of circ PTBP3 is closely related to the T stage,N stage,TNM stage and overall survival rate of colorectal cancer patients.Further functional experiments showed that after interfering with circ PTBP3,colorectal cancer cells SW620 and HCT116 can proliferate,migrate and invasively weaken in vitro.Western blotting showed that circ PTBP3 may be able to regulate the EGFR signaling pathway,and interfere with circ PTBP3 to reduce the protein levels of EGFR and MAP3K1.Conclusion:circ PTBP3 is highly expressed in colorectal cancer tissues and cell lines,and the high expression of circ PTBP3 is related to the T stage,N stage,TNM stage and overall survival rate of colorectal cancer.circ PTBP3 promotes the proliferation,migration,and invasion of colorectal cancer cell lines HCT116 and SW620.circ PTBP3 may promotes the progression of colorectal cancer through the EGFR/MAPK signaling pathway.
