| Objective:To investigate the correlations of lymphocyte antigen 96(LY96)to clinically characterize and prognosis in gliomas,and further analyze the correlation with between LY96 expression and immune infiltration in glioma.Methods:A retrospective analysis explored clinically characterize and prognosis landscape of LY96 in gliomas based on The Cancer Genome Atlas(TCGA)and Chinese Glioma Genome Atlas(CGGA)datasets.LY96 protein-protein interaction network was investigated via STRING,function enrichment analysis was used to explore the potential molecular functions and pathways.We then investigated the correlation between LY96 expression and levels oftumor-infiltrating immune cells and their gene markers based on TIMER.Results:Compared with the low grade glioma(LGG)group,LY96 was highly expressed in glioblastoma multiforme(GBM)(P<0.001),and followed the World Health Organization(WHO)classification of glioma Increased,the expression level of LY96 also gradually increased(P<0.001).There was no statistical difference between gender and the expression of LY96(P>0.05);LY96 was significantly overexpressed in elderly(≥ 60years)patients(P<0.001);the expression of LY96 in IDH1 wild type was significantly higher than IDH1 mutant type(P<0.001);the expression of LY96 in 1p/19qno co-deletion was significantly higher than co-deletion group(P<0.001).In the study of all gliomas,GBM and LGG,the overexpression of LY96 was correlated with poorer over survival(all P<0.05).The multivariate Cox regression model identified that LY96 expression,elderly(≥60years)and tumor grade(GBM vs LGG)were independent risk factors for the prognosis landscape of glioma.Included 31 LY96 interacted proteins.via STRING,aggregation index 0.842(P<0.01);biological functions focused on immune system process,regulation of response to external stimulus,protein binding and signaling receptor binding,etc.Cellular component focused on membrane,membrane-bounded organelle and intracellular organelle.Signal pathways concentrated on Toll-like receptor signaling pathway and NF-kapa B signaling pathway.The expression of LY96 was negatively associated with tumor purity in GBM and LGG.Interestingly,LY96 expression was positively correlated with most gene markers of tumor-infiltrating immune cells in GBM and LGG.Conclusion:These findings suggest that LY96 expression could be an independent prognostic factor to predicte glioma patient outcomes,which is associated with immune infiltration,LY96 could be a potential target for immunotherapy of glioma. |
