Objective:Through observing the changes of the key signal molecules and inflammatory factors in transforming growth factor-β1(TGF-β1)/Smads signal pathway,this study explored the mechanism of Bushen Tongluo method to improve IPF,in order to provide new ideas and methods for clinical treatment of IPF.Methods:40 SPF male SD rats(150±30)g were randomly divided into five groups:normal group,model group,low dose group,high dose group and control group.Except for the normal group,the other groups were induced by bleomycin to make the rat model of idiopathic pulmonary fibrosis,Three days after the establishment of the model,the normal group and the model group were given normal saline,the high and low dose groups were given 14.2 g·kg-1and 7.1 g·kg-1respectively,and the control group was given 4.58g·kg-1hydrocortisone sodium succinate once a day intraperitoneal injection.After 28 days of continuous administration,the rats were decapitated and killed.The levels of serum HYP,HA,LN,IL-1,IL-6,TNF-α,TGF-β1,Smad2,Smad3,Smad7 protein and m RNA were measured.Results:1.Comparison of pathological changes of lung tissue in each groupHE staining of lung tissue:Compared with the normal group,the alveolar structure of the model group was disordered,the alveoli were atresia and collapse,some of them fused into bullae,and there were inflammatory cell infiltration in the alveoli cavity and interstitium.Compared with the model group,the degree of alveolitis in the high dose group,the low dose group and the positive drug control group decreased.Masson staining of lung tissue:compared with the normal group,the alveolar structure of the model group was disordered,and a large number of collagen fibers were proliferated and deposited in the lung interstitium.Compared with the model group,the degree of alveolitis in the high dose group,the low dose group and the positive drug control group decreased.2.Comparison of the contents of HYP,HA and LN in each group of ratsCompared with the normal group,the content of HYP,HA and LN in the model group increased(P<0.05 or P<0.01);compared with the model group,the content of HYP,HA and LN in the high dose group of Bushen Tongluo decreased(P<0.05 or P<0.01),and the content of LN in the low dose group of Bushen Tongluo decreased(P<0.01).3.Comparison of inflammatory factors in lung tissue of rats in each groupCompared with the normal group,the levels of TNF-α,IL-1 and IL-6 in the model group were all increased(P<0.01).Compared with the model group,the content of TNF-α,IL-1 and IL-6 in Bushen Tongluo high dose group decreased(P<0.05 or P<0.01),The content of TNF-α,IL-1 and IL-6 decreased in the positive drug control group(P<0.05 or P<0.01).4.Expression of TGF-β1/Smads pathway related indexesCompared with the normal group,the expression of TGF-β1,Smad2and Smad3 in the model group increased(P<0.01),The expression of Smad7decreased(P<0.01),The m RNA expression of TGF-β1,Smad2 and Smad3increased(P<0.05 or P<0.01)The expression of Smad7 m RNA decreased(P<0.01).Compared with the model group,the expression of TGF-β1,Smad2,Smad3 decreased(P<0.05),Smad7 increased(P<0.01),TGF-β1,Smad2,Smad3 m RNA decreased(P<0.05),Smad7 m RNA increased(P<0.05).The expression of TGF-β1 and Smad3 decreased in the low dose group(P<0.05 or P<0.01)The expression of Smad7 m RNA increased(P<0.05).The expression of TGF-β1 and Smad3 decreased in the positive drug control group(P<0.05 or P<0.01),The expression of Smad7 increased(P<0.01)The expression of TGF-β1 and Smad2 m RNA decreased(P<0.05)The expression of Smad7 m RNA increased(P<0.05)。Conclusion:Bushen Tongluo Decoction can reduce the content of serum markers,improve the pathological morphology of pulmonary fibrosis and delay the process of idiopathic pulmonary fibrosis in rats with pulmonary fibrosis.The mechanism may be related to the regulation of TGF-β1/Smads signal pathway and the decrease of inflammatory cytokines in lung tissue.
