| Objective:In this experiment,the effects of Tongli Sanjiao Qingre Lishi prescription on transforming growth factor-β1(TGF-β1),β1,3galactosyltransferase(C1Gal T1;encoded by C1GALT1)and core 1 β3-Gal-T soerific molecular chaperone(C1Gal T1C1;encoded by C1GALT1C1)in renal tissue of IgA nephropathy(IgAN)rats were observed.To provide theoretical basis for clinical application of Tongli Sanjiao and Qingre Lishi formula in the treatment of IgAN.Methods:48 clean male SD rats were selected.After one week of adaptive feeding,the ones with negative protein after urinalysis entered the experiment and were randomly divided into 4 groups: normal group(group N),model group(group M),and benazepril group(group X)and Chinese medicine group(group Z).The bovine serum albumin + lipopolysaccharide +carbon tetrachloride + castor oil immune induction method was applied to replicate the experimental IgAN rat model.After the model was successfully made,the rats were given the corresponding treatment medicines and distilled water for intragastric administration,then the 24-hour urine volumes of rats in each group were collected on the 9th and 13 th weekends to do 24h-UTP test.After 14 weeks of experiment,the rats were executed,and femoral arterial blood and kidney tissues were reserved.Biochemical indicators such as serum creatinine(Cr),blood urea nitrogen(BUN),albumin(ALB),total cholesterol(TC),and triglyceride(TG),and the content of TGF-β1 in the blood were measured.In addition,the local pathological changes in the kidney and the expression of TGF-β1,C1 Gal T1,C1 Gal T1C1 protein and genes in kidney tissues were observed.Result:1.In the whole process of the experiment,the rats in group N were in good condition as a whole;the other three groups showed different degrees of abnormal performance in hair color,diet,spirit,activity,etc.After the corresponding treatment,the overall situation of group X and group Z was better than that of group M.2.Blood biochemical indexes:(1)Tc: compared with group N,the content of TC in serum of group M,X and Z was higher(P < 0.05);compared with group M,the content of TC in group X and Z was lower(P < 0.05).(2)There was no significant difference in serum Cr,BUN,ALB and TG between the four groups(P > 0.05).3.The results of 24h-UTP in N group were negative in each period,and the other three groups were increased in varying degrees.Compared with group M,the results of group X and Z were lower after corresponding treatment(P < 0.05),and there was no statistical difference between group X and Z(P >0.05).4.The content of TGF-β1 in the serum of group M,X and Z was higher than that of group N(P < 0.05),and the content of TGF-β 1 in the serum of group X and Z was lower than that of group M(P < 0.05).There was no significant difference between group X and Z(P > 0.05).5.The renal tissue was observed by light,immunofluorescence and electron microscopy.There was no renal pathological change in group N,the most obvious pathological change was happened to group M,and the renal pathological changes in group X and Z were significantly less than those in group M.6.The expression of TGF-β1 and m RNA in renal tissue of rats were observed by immunohistochemistry or RT-PCR.The results showed that the expression of TGF-β1 and m RNA in group M,X and Z was significantly higher than that in group N(P < 0.05),and the expression of TGF-β1 and m RNA in group X and Z was lower than that in group M(P < 0.05).There was no significant difference between groups X and Z(P > 0.05).7.The expression of C1 Gal T1,C1 Gal T1C1 and m RNA in renal tissue of rats were observed by Weatern blot or RT-PCR.It was found that the expression of group M,X and Zwas significantly lower than that of group N(P < 0.05),and the expression of group X and Z was significantly higher than that of group M(P < 0.05).There was no significant difference betweengroup X and Z(P > 0.05).Conclusion:1.Tongli Sanjiao Qingre Lishi prescription can significantly reduce the24-hour urinary protein quantification of IgA nephropathy rats,and its effect is equivalent to benazepril hydrochloride.2.Tongli Sanjiao Qingre Lishi prescription can significantly reduce the pathological damage of renal tissue in IgA nephropathy rats,and its effect is equivalent to benazepril hydrochloride.3.Tongli Sanjiao Qingre Lishi prescription can significantly reduce the expression of TGF-β1 and enhance the expression of C1 Gal T1 and C1 Gal T1C1 in renal tissue of IgA nephropathy rats,and the effect is equivalent to benazepril hydrochloride. |
