| ObjectiveGastric cancer is a malignant tumor disease with a high morbidity and a high mortality rate in the world.The situation is particularly serious in China.The diagnosis and treatment of gastric cancer is related to the stage of the disease.Early diagnosis can greatly improve the curative effect,but it lacks ideal tumor markers.Circular RNA is a new type of endogenous non-coding RNA.Thanks to its unique circular structure,it has the characteristics of high stability,high conservation and high abundance,and plays an important role in tumor diseases.Therefore,this study takes gastric cancer as the research object,circular RNA as a solution,and uses molecular biology techniques to explore a tumor biomarker that is applied to the diagnosis and treatment of gastric cancer.Methods1.Using human circular RNA gene chip,three pairs of gastric cancer patient tissues were tested.Genes differentially expressed in gastric cancer and normal tissues adjacent to the cancer were screened,and gastric cancer-related circular RNAs were selected as biomarkers.2.100 pairs of gastric cancer tissues and normal tissues adjacent in gastric cancer patients were subjected to RNA extraction,reverse transcription to c DNA.The expression of hsa_circ_0005556 of 100 paired GC tissues and adjacent normal tissues was detected using quantitative reverse transcription-polymerase chain reaction(q RT-PCR).3.Establish receiver operating characteristic curve(ROC)curve and evaluate the value of hsa_circ_0005556 in the diagnosis of gastric cancer.4.Combine the clinical pathological data and survival data to analyze the value of hsa_circ_0005556 in the prognosis of gastric cancer patients.5.Use bioinformatics tools to explore potential biological pathways and functions of hsa_circ_0005556.Results1.According to q RT-PCR test results,the expression level of hsa_circ_0005556 in gastric cancer tissues of gastric cancer patients was significantly lower than that of normal adjacent tissues(n = 100,p <0.001).2.The area under the ROC curve(AUC)of Hsa_circ_0005556 is 0.773.When the cut-off value is 11.74,the sensitivity and specificity are 64% and 82%,respectively.3.The expression of hsa_circ_0005556 in gastric cancer tissues is related to some clinicopathological factors,such as tumor differentiation(p = 0.001),lymphatic metastasis(p =0.039)and TNM stage(p = 0.013);but not related to age,gender,tumor diameter,degree of invasion,distant metastasis,blood carcinoembryonic antigen,blood glucose antigen 199.4.The expression level of hsa_circ_0005556 in gastric cancer tissue can reflect the survival level.The overall survival(OS)of patients with low expression group hsa_circ_0005556 is significantly worse than that of high expression group(p = 0.047).ConclusionsThis study explored and verified the clinical value of hsa_circ_0005556 in the diagnosis and treatment of gastric cancer.Hsa_circ_0005556 can be used as a biomarker of gastric cancer,which is helpful for diagnosis and prognosis.At the same time,hsa_circ_0005556 can assist in determining the indications for early gastric cancer endoscopic treatment,and promote the application of minimally invasive treatment of early gastric cancer. |
