Objective:Selection of chemical radiculitis rat model to simulate lumbar disc herniation.To observe the effect of finger-pressure Qihai,Yanglingquan and Kunlun points on the behavior,morphology and serum IL-1 β,IL-6,TNF-α,NGF,NT-3 of lumbar disc herniation.This study aims to explore the mechanism of the treatment of lumbar disc herniation mainly by finger Qihai,and to provide new ideas and objective data support for clinical treatment.Methods:By using SPSS 22.0 statistical software,forty male SPF rats(180 ± 20g)were randomly divided into normal group(8 rats),sham operated group(8 rats)and modeling group(24 rats).In the modeling group,the skin was cut 0.3cm to the left of the midpoint between the right iliac crest and ischial tubercle.Separating the muscles on both sides and exposing nerve root fully.The circular filter paper soaked with 19% formalin liquid was placed under the nerve root armpit for modeling.Rats in normal group have no operations.In the sham operation group,only circular filter paper was not placed.On the third day of modeling,the Siegal nerve function score was used to evaluate the model rats.Then SD rats were randomly divided into model group(8 rats)and model finger-pressure group(8 rats)by SPSS 22.0.Finally,there were 8 rats in normal group,8 rats in sham operated group,8rats in model group and 8 rats in model finger-pressure group.Recording the Siegal neural function scores and the paw withdrawal latency(PWL).Blood was collected from the fundus artery of each group of rats.Serum was separated and stored in-80℃ refrigerator.On the fourth day of modeling,the normal group,the sham operation group and the model group were only fixed and grasping,while the pointer group was treated with the pointer once a day,a total of 14 times.After treatment,the behavior of rats was detected.Blood was taken from abdominal aorta.Serum was separated and stored in refrigerator at-80 ℃.The dorsal root ganglion(DRG)was removed for Hematoxylin Eosin(HE)staining.The concentrations of TNF-α,IL-1 β,IL-6,NT-3 and NGF in serum samples were detected by enzyme-linked immunosorbent assay(ELASE).Results:1.Behavioral evaluationThe results of the nerve function scores:Before treatment,the neurological function scores of the finger-pressure and the model group were significantly higher than those of the normal group and the sham operation group(P < 0.05).There was no significant difference between the finger-pressure and the model group(P > 0.05),and there was no significant difference between the normal group and the sham operation group(P > 0.05).After treatment,the nerve function scores of the finger-pressure and the model group were significantly lower than those before treatment(P < 0.05);Compared between groups,the nerve function score of the finger-pressure was significantly lower than that of the model group(P< 0.05).The results of paw withdrawal latency due to hot pain(PWL): Before treatment,PWL in finger-pressure and model group was significantly higher than that in normal group and sham operation group(P < 0.05),there was no significant difference between finger-pressure and model group(P > 0.05),and there was no significant difference between normal group and sham operation group(P > 0.05).After treatment,PWL in finger-pressure was significantly lower than that before treatment(P < 0.05).But compared before treatment,there was no significant difference in normal group,sham operation group and model group(P > 0.05).Compared between groups,PWL in finger-pressure was significantly lower than that in model group(P < 0.05).2.Results of serum TNF – α,IL-1 β,IL-6,NT-3 and NGF concentrationsBefore treatment,the serum levels of TNF-α,IL-1 β and IL-6 in the finger-pressure and model group were significantly higher than those in the normal group and the sham operation group(P < 0.05).There was no significant difference between the finger-pressure and the model group(P > 0.05),and there was no significant difference between the normal group and the sham operation group(P > 0.05).After treatment,the concentrations of TNF-α,IL-1 β and IL-6 in the finger-pressure were significantly lower than those before treatment(P < 0.05).And compared before treatment,there were no significant differences in the normal group,the sham operation group and the model group(P > 0.05);Compared between groups,the concentrations of TNF-α,IL-1 β and IL-6 in the finger-pressure were significantly lower than those in the model group(P < 0.05).Before treatment,the serum concentrations of NGF and NT-3 in the finger-pressure and the model group were significantly higher than those in the normal group and the sham operation group(P < 0.05).There was no significant difference between the finger-pressure and the model group(P > 0.05),and there was no significant difference between the normal group and the sham operation group(P > 0.05).After treatment,the concentration of NGF in the finger-pressure was significantly lower than that before treatment(P < 0.05),and the concentration of NT-3 was significantly higher than that before treatment.There was no significant difference in the concentration of NGF and NT-3 in the normal group,the sham operation group and the model group compared with before treatment(P > 0.05);Compared between groups,the NGF concentration in the finger-pressure was significantly lower than that in the model group(P < 0.05),and the concentration of NT-3 was significantly higher than that in the model group(P < 0.05).3.HE staining results of DRG in ratsAfter 14 days of treatment,under 400 times of microscope,the neurons in the ganglion of the model group were swollen,the cytoplasm was loose and swollen obviously,the number of Nissl bodies in the cytoplasm was less or disappeared,some of them were vacuolated and denatured,a few of them were pyknotic,necrotic and replaced by glial cells.In the model finger-pressure group,the degree of cytoplasmic looseness,vacuolar degeneration and edema of neurons in the ganglion was significantly reduced compared with the model group,and the cytoplasmic Nissl bodies recovered.Conclusion:1.The results showed that finger-pressure Qihai,Yanglingquan,Kunlun points could promote the recovery of Siegal neural function scores and PWL,and repair the motor and sensory functions of injured nerves.2.Finger-pressure Qihai,Yanglingquan,Kunlun points can reduce the level of TNF-α,IL-1β,IL-6,NGF,raise the level of NT-3 in serum,then weaken the inflammatory response and repair the damaged nerve.3.Finger-pressure Qihai,Yanglingquan,Kunlun points can reduce the edema of DRG neurons,increase the number of Nissl bodies and promote the repair of DRG tissues.
