| Kudzu(Puerariae Lobatae Radix and Puerariae Thomsonii Radix)has a long history as a traditional Chinese medicine.It has a variety of biologically active ingredients such as lowering blood pressure and blood lipids.As edible traditional Chinese medicine,it has high application value and is widely used in drug development,health food development and common food development.Kudzu has higher safety,but it had been reported to have a certain degree of hepatotoxicity.At the same time,Kudzu is rich in isoflavones and has estrogen-like effects,whether there are any safety or adverse effects on specific populations remains unknown.In this study,adolescent SD rats eaten Kudzu in high doses and 30-day feeding method was combined with modern metabolomics techniques to evaluate safety of organs and tissues such as liver,spleen,kidney,different sections of the intestine and reproductive organs of adolescent SD rats.1.safety study of adolescent SD rats of KudzuIn the safety study of normal adolescent SD rats,Kudzu has different effects on adolescent female and male SD rats.There were no abnormalities in the blood routine of different groups of taking kudzu.Among the effects on liver function,Puerariae Lobatae Radix had a certain effect on the serum biochemical indexs ALP,ALT and TP of liver function in female SD rats,but the biochemical indexs of TBIL,DBIL,TBA,AST,Lp(a),ADA,AFU and ALB were no abnormalities.Puerariae Thomsonii Radix had a certain effect on the serum biochemical indexes ALT and TBA of liver function in male SD rats,but the biochemical indexs of ALP,TBIL,DBIL,AST,TP,Lp(a),ADA,AFU,ALB were no abnormalities.Puerariae Lobatae Radix had no significant effect on the biochemical indexes of liver function in male SD rats,while Puerariae Thomsonii Radix had no significant effect on the biochemical indexes of liver function in female SD rats.Moreover,no pathological changes were found in the livers of SD rats.The results showed that kudzu had a certain effect on liver function in SD rats,however,the combination of oxidative damage indexs and pathological results of male and female SD rats showed that no organic lesion was found in the livers and no obvious hepatotoxicity was found.Among the effects on renal function,Kudzu had no significant effect on the serum biochemical indexes BUN,CRE and UA of renal function in male and female SD rats,it had a certain effects on SOD,MDA and GSH in the kidneys of male and female SD rats,but no obvious pathological changes were found in the pathological sections.It suggested that Kudzu had adverse effects on renal function in SD rats,but no obvious renal toxicity was found.There were no pathological changes in the duodenum,jejunum,ileum and colon tissue,it suggested that Kudzu had no obvious toxicity to the intestine.Among the effects on sex organs,Puerariae Lobatae Radix and Puerariae Thomsonii Radix can increase the estradiol level in the serum of male and female SD rats,the level of aromatase in the testis was not significantly increased and no oxidative damage occurred,combined with the testicular tissue morphology and testicular organ coefficients,it suggested that the intake of kudzu did not affect the development of male sexual organs under the experimental conditions.Combined with histopathological examination of ovary,uterus and breast,it was suggested that the intake of kudzu did not significantly promote the precocity of female organs in this experimental condition.2.safety study of adolescent SD rats of Kudzu based on serum metabolomicsIn the safety study of normal adolescent SD rats of Kudzu based on serum metabolomics,7 to 21 different metabolites were screened.The affected metabolic pathways mainly include phenylalanine,tyrosine and tryptophan biosynthesis,phenylalanine metabolism,steroid biosynthesis,aminoacyl-t RNA biosynthesis,steroid hormone biosynthesis.Combined with-log(P)> 2 and Pathway impact > 0.1 shows that Kudzu had a greater effect on the phenylalanine,tyrosine and tryptophan biosynthesis or phenylalanine metabolism,mainly manifested by inhibiting the conversion of phenylalanine to tyrosine and tryptophan or inhibiting the metabolism of phenylalanine.3.Research on the development and quality evaluation of related products of Puerariae Lobatae Radix juiceThe prescription "Gegen Yin" was determined by the prescription screening in ancient books combined with the results of metabolomics research.After the development of Puerariae Lobatae Radix juice related products,qualitative identification method and content determination method were established.The product parameters such as p H value,soluble solids,relative density,total flavonoid content,puerarin content and daidzein content were collected to establish product quality standards,completed the development of Puerariae Lobatae Radix juice related product. |
