| Objective At present,it is known that interleukin-13(IL-13)plays an important role in the cognitive function as an anti-inflammatory cytokine;brain-derived neurotrophic factor(BDNF)protects neurons through various ways,which is conducive to the formation of learning and memory;β-amyloid is found in the brain of a large number of patients with cognitive impairment The deposition of β-protein,a β)results in the damage of nerve cells and the decrease of learning and memory ability.The expression of IL-13,BDNF and a β are all sensitive to the change of sleep,but there are few reports about circadian rhythm disorder.Therefore,the purpose of this experiment is to explore the changes of IL-13,BDNF and a β in the hippocampus of mice in the process of circadian rhythm disorder under the condition of continuous light,to help further explore the possible relationship and interaction mechanism between them,and to explore the changes and roles of IL-13 in circadian rhythm disorder hippocampus.Method1.C57 BL / 6J male mice were screened for 7-8 weeks.Based on this,they were divided into normal circadian rhythms(NCR),circadian rhythms disorder(CRD)group and as1517499 + CRD group with 10 mice in each group.NCR group was killed 21 days after feeding in the same cage under normal light conditions,CRD group was killed 21 days after continuous light conditions,as1517499 + CRD group needed to use as1517499 for intraperitoneal injection,and was killed 21 days after continuous light conditions.2.The expression levels of IL-13,BDNF and a β in the hippocampus of mice in each group were observed by ELISA.Result1.The results of ELISA showed that the expression of IL-13 in hippocampus of CRD group was higher than that of NCR group,the difference was statistically significant(P < 0.05);the expression of IL-13 in as1517499 + CRD group was higher than that of NCR group,the difference was statistically significant(P < 0.05);the expression of IL-13 in as1517499 + CRD group had no significant change compared with that of CRD group(P > 0.05).2.The results of ELISA showed that the expression of BDNF in hippocampus of CRD group was higher than that of NCR group,but the difference was not statistically significant(P > 0.05);the expression of BDNF in as1517499 + CRD group was lower than that of NCR group,the difference was not statistically significant(P > 0.05);the expression of BDNF in as1517499 + CRD group was lower than that of CRD group,and the difference was not statistically significant(P > 0.05).3.Pearson correlation analysis showed that the levels of IL-13 and BDNF in hippocampus of circadian rhythm disorder mice were positively correlated,and the correlation coefficient was r = 0.687,P < 0.01.4.The results of ELISA showed that there was no significant difference in the expression of a β in CRD group compared with NCR group(P > 0.05);there was no significant difference in the expression of a β in as1517499 + CRD group compared with NCR group(P > 0.05);there was no significant difference in the expression of aβ in as1517499 + CRD group compared with CRD group(P > 0.05).There was no significant correlation between a β level and IL-13,BDNF.Conclusion1.The disorder of circadian rhythm increased the level of IL-13 in mouse hippocampus,suggesting that the disorder of circadian rhythm stimulated the expression of anti-inflammatory factor IL-13.2.The expression of BDNF in hippocampus of mice with circadian rhythm disorder was higher than that of mice with normal light condition,but there was no statistical difference.3.In the course of circadian rhythm disorder,the expression of IL-13 has a positive correlation with BDNF,suggesting that IL-13 has a regulatory effect on BDNF.4.There was no significant difference in the expression of a β in the hippocampus of circadian rhythm disorder mice compared with the normal baseline level. |
