In practice,forensic toxicology analysis is often faced with the extraction of trace amounts of drugs from complex matrix,the identification of species and the measurement of content.In forensic practice,it is a difficult problem for forensic personnel to accurately determine the nature and target of toxic substances.Based on legal medical toxicology,using Ultra Performance Liquid Chromatography-Mass Spectrometry(UPLC-MS/MS)method,this paper provides ideas and methods to solve such a kind of problem in judicial practice,that is,the suspect of amphetamines positive cases claims to take prescription drug,selegiline,and denies taking drugs.In order to better apply scientific evidence to judicial practice,this paper explores the attributes of scientific evidence.In the experimental part of this paper,the effects of different procedures were investigated by using UPLC-MS/MS.Finally,two detection methods were established:(1)Oasis PRiME HLB solid phase extraction column was used to treat the substrate,and the samples were analyzed by Waters XeVo TQS UPLC-MS/MS.The chromatographic column was ACQUITY UPLC HSS C18(150×2.1mm,1.8μm).The organic phase A was acetonitrile,the aqueous phase B was 5mmol/L ammonium formate formic acid aqueous solution(pH=3),the flow rate was 0.4ml/min,gradient elution,and the injection volume was 3μL,electrospray ionization(ESI),positive ion scanning and mass spectrometry multiple reaction detection.The target peaks of selegiline were detected within 4 min.(2)Oasis PRiME MCX solid phase extraction column was used to treat the substrate,and the samples were analyzed by LC-30A-QTRAP5500 UPLC-MS/MS.The column was Phenomenex Lux AMP(150×3.0mm,3μm).The organic phase A was methanol,the aqueous phase B was 5mmol/L ammonium bicarbonate solution(adjusted with ammonia water pH=11),the flow rate was 0.35 mL/min,gradient elution,the injection volume was 2μL,electrospray ionization(ESI),positive ion scanning,mass spectrometry multiple reaction detection,the target substances are separated within 20min.The method was applied to the practical samples.This paper also makes a theoretical research on the attribute of scientific evidence.As a kind of evidence,scientific evidence also has four attributes: relevancy,admissibility,probative force and credibility.Judges,lawyers and other judicial personnel should have a better understanding of the four attributes of scientific evidence.To grasp the basic attribute of relevance;to use the likelihood ratio to explain the evidence,not blindly credulous of scientific evidence;do not presuppose the natural probative force of scientific evidence ang keep procedural justice;to improve the scientific of test means;and ensure the credibility of scientific evidence.The method established in this paper is fast,easy to operate,high sensitivity,and can be applied to the laboratory detection of poisons and drugs.The attribute of scientific evidence can provide some reference for judicial practice,but the experimental detection method can be further optimized,and the theoretical research of scientific evidence can be further deepened.
