| Background:At present,cancer has become a major factor affecting the public health.Pancreatic cancer is the most common pancreatic tumor with a high degree of malignancy.Due to its low early diagnosis rate and high mortality,it is prone to metastasis with a low rate of survival.Therefore,it is of great significance to find new methods for the diagnosis and treatment of pancreatic cancer.In recent years,some studies have shown that F-box protein is a kind of protein family containing F-box domain.The F-box domain,located at the N-terminal of F-box protein,connect with SKP1 and Cullin proteins to form SCF(SKP1-CUL1-F-box)E3 ubiquitin ligase complex.The C-terminal of F-box protein is mainly involved in the recognition of specific substrate proteins,the function of F-box protein is mainly related to tumor development,cell cycle regulation,cell metabolism and other processes.FBXO45,as a member of the protein family,participates in the regulation of the evolution and progression of malignant tumors,however,there is no relevant report in pancreatic cancer.Therefore,aim to explore the expression of FBXO45 in pancreatic cancer and its effect on proliferation and invasion of pancreatic cancer cells,this study hopes to explore the effect of FBXO45 on the tumor growth of mice.In this way,this study can further clarify the molecular mechanism in regards to the occurrence and development of pancreatic cancer,and to provide more theoretical and experimental references for future studies on potential molecular targets of pancreatic cancer.Purposes:We investigate the expression of FBXO45 in pancreatic cancer and its effect on malignant biological functions of pancreatic cancer cells.Methods:Pancreatic cancer cells Panc-1 and Patu-8988 were cultured in vitro.FBXO45 plasmid and small interference fragments were transfected into the above cells.After the transfection,the effects of FBXO45 overexpression and knockout were measured by Western blotting.The malignant biological behaviors of pancreatic cancer cells were observed by cell proliferation assay,wound healing assay and Transwell assay.The immunohistochemistry assay was used to detect the expression of FBXO45 in human pancreatic cancer tissues and its relationship with clinical prognosis.To observe the effect of FBXO45 on tumor growth in nude mice,pancreatic cancer cell lines with stable transfection of FBXO45 were constructed and subcutaneous tumor model was established.Results: After the transfection of FBXO45 plasmid,the proliferation,migration and invasion of pancreatic cancer cells were significantly enhanced.Down-regulation of FBXO45 expression significantly reduced the proliferation,migration and invasion of pancreatic cancer cells.Immunohistochemical results showed that,compared with adjacent tissues,the expression of FBXO45 was significantly increased in human pancreatic cancer tissues.Results of survival curve analysis showed that patients with high FBXO45 staining score had a lower survival rate than those with low FBXO45 staining score.The results of animal experiments revealed that the subcutaneous tumors of nude mice with FBXO45 overexpression were significantly larger than those of the control group.Conclusions: FBXO45 can enhance the proliferation ability of pancreatic cancer cells and promote their migration and invasion.The expression of FBXO45 was negatively correlated with better survival of pancreatic cancer patients.Moreover FBXO45 promoted the growth of subcutaneous tumors in mice. |
