In this paper,Hyp is used as the experimental material,and the multi-omics joint analysis method is used to comprehensively explore the regulation and mechanism of Hyp on lipid metabolism in high-fat mice.The specific research results are as follows:1.By measuring the changes of Hyp on the body weight,blood lipid level,liver tissue morphology and structure of high-fat mice,the effect of Hyp on lipid metabolism in mice was explored.The results show that Hyp can significantly reduce the body weight and the levels of TG,TC,and LDL in high-fat mice,so that the external morphology and internal structure of liver tissue can be effectively restored.2.The Illumina platform was used to determine and analyze the effect of high-fat mice.As a result,the abundance of AF12,Ruminococcus,etc.was significantly increased and the abundance of Sutterella,etc.was significantly decreased as compared with the HFD group.This indicates that Hyp can reduce the abundance of pathogenic bacteria such as Sutterella by increasing the abundance of beneficial bacteria such as AF12.3.Using LC-MS technology to determine and analyze the effect of Hyp on metabolites in feces of high-fat mice.The results showed that compared with the HFD group,a total of 18 significantly different metabolites such as retinoic acid were identified.KEGG enrichment analysis showed that the pathways that have significant effects on mouse metabolism are taurine and taurine metabolic pathways and retinol metabolic pathways.4.The Illumina platform was used to determine and analyze the effect of Hyp on intestinal gene expression in high-fat mice.The results showed that a total of 267 genes with significant differences were screened compared to the HFD group.Annotated analysis using KEGG,chemical carcinogenesis and retinol metabolic pathways have a profound effect on mouse metabolism.5.Through the joint analysis of different intestinal flora,different metabolites and different genes,explore the mechanism of Hyp regulating lipid metabolism in mice.The results of combined analysis of intestinal differential flora and differential metabolites show that Hyp can adjust the lipid metabolism of high-fat mice by increasing the abundance of AF12,reducing the abundance of unidentified_Desulfovibrionaceae,increasing the content of retinoic acid,and improving fat deposition.The combined analysis of differential metabolites and differential genes showed that Hyp can reduce the expression of Cyp1a2,Ugt1a6 b and other genes,increase the content of retinoic acid,and regulate lipid metabolism in high-fat mice.In summary,Hyp can reduce the expression of Cyp1a2,Ugt1a6 b and other genes by changing the abundance of intestinal flora such as AF12 and unidentified_Desulfovibrionaceae,increase the content of retinoic acid,and regulate lipid metabolism in high-fat mice.
