| Objective: Irisin is a new hormone secreted by skeletal muscle,it has been found that it is closely related to endocrine and metabolic diseases,cardiovascular diseases,tumors,bone metabolism and so on.To explore the effect of irisin on osteogenic differentiation of bone marrow mesenchymal stem cells and its possible mechanism is helpful to provide new ideas for fracture healing.Materials and methods: Mouse bone marrow mesenchymal stem cells were extracted and cultured.The range of cell proliferation activity of irisin was verified by cck-8 test.Irisin was cultured with different concentrations of irisin.The effect of irisin on the proliferation and osteogenic differentiation of BMSCs was verified by alizarin red staining and ALP activity detection.The activity of Wnt/ β-catenin signal pathway stimulated by osteogenic associated protein and irisin was analyzed by Western blot technique.Results: Irisin could promote the proliferation of bone marrow mesenchymal stem cells and the formation of mineralized nodules.Under the stimulation of Irisin,Runx2,OCN,ALP and osteocalcin were significantly up-regulated,and the activity of Wnt/ β-catenin signal pathway was also significantly enhanced.Conclusion: Irisin can promote the proliferation and osteoblast differentiation of bone marrow mesenchymal stem cells through Wnt/ β-catenin signal pathway. |
