Objective:To study the effect and mechanism of peimine,a major effective component in herbal Fritillaria thunbergii,on proliferation,apoptosis and cell cycle of breast cancer MCF-7 cellsMethods:MCF-7 cells in good condition were taken into two groups.One was labeled as the experimental group and added with peimine solution in the culture medium,the other was labeled as the control group and added with DMSO solution.CCK8 kit was used to detect the cell proliferation ability of the experimental group and the control group.Annexin V and PI were used to label the cells of experimental group and control group,and the apoptosis of each group was detected by flow cytometry;Cell cycle staining kit was used to label the cells in the experimental group and the control group,and the percentage of cells in each phase of each group was detected by flow cytometry;The expressions of p21,cyclin D1,cyclin A and cyclin E were detected by Western blot and fluorescence quantitative PCR.Results:Compared with the control group,the proliferation ability of the experimental group was inhibited in a dose-dependent manner(p<0.01),and the apoptosis of the experimental group was increased compared with the control group(p<0.05);Cell cycle analysis showed that the proportion of G1 phase cells in the experimental group was higher than that in the control group(p<0.05).Meanwhile,Western blot and fluorescence quantitative PCR showed that the cyclin D1 in the experimental group decreased while the p21 protein increased significantly in the experimental group(p<0.001).Conclusion:Peimine can inhibit the proliferation of MCF-7 cells and increase the apoptosis of MCF-7 cells.The cell can be arrested in G1 phase of cell cycle by peimine,the possible mechanism of which is that peimine can promote the expression of p21 and reduce the expression of cyclin D1.These results provide a basis for the treatment of breast cancer and drug development.
