| Objective:To investigate the protective effect and mechanism of eighteen flavor myrobalan diuretic pills on diabetic nephropathy(DN)in rats.Methods: Thirty male SD rats,10 rats were randomly selected as the control group,and the remaining rats were treated with high glucose and high fat diet + STZ injection to replicate the DN rat model.The successful rats were divided into model group and Tibetan medicine group by random number method.Lavage 2 weeks in a row,The urine of rats for 24 hours was collected,and the blood of abdominal aorta was collected for detection the Serum creatinine(SCR),blood urea nitrogen(BUN),uric acid(UA),cystatin C(Cys-C),β2 microglobulin(β2-MG),retinol binding protein(RBP),triglyceride(TG),total cholesterol(TC),high density lipoprotein(HDL),low density lipoprotein(LDL),fructosamine(FRA).The pathological changes of renal tissue were observed under light microscope and electron microscope;Immunohistochemistry and Western blot were used to detect the expressions of RAGE、NF-κB P65、TNF-α、IL-6、IL-1β in kidney tissues.Results:1.Comparison of general condition of rats in each group: the rats in the control group were obese and the hair was shiny and sensitive.Rats in model group were emaciated,with yellow hair and visible shedding,arched back up,low energy,slow reaction and polyphagia,polyphagia and polyuria.The general condition of rats in the Tibetan medicine group was significantly improved compared with that in the model group.2.Changes in renal function of rats in each group:Compared with the control group,24 h PRO、Scr、BUN、UA、Cys-C、β2-MG、RBP in model group was significantly increased(p<0.05 or p<0.01),Compared with model group,24 h PRO、Scr、BUN、β2-MG in Tibetan medicine group was significantly decreased(p<0.05 or p<0.01)there was no significant difference in UA、Cys-C、RBP(p>0.05).3.Changes of FRA in each group: Compared with the control group,FRA in the model group was significantly increased(p<0.01),the FRA in Tibetan medicine group was significantly lower than that in model group,The difference was statistically significant(p<0.05).4.Changes of blood lipid in each group: Compared with the control group,TG、TC and LDL in model group were significantly increased(p<0.01),HDL was significantly decreased(p<0.01),compared with the model group,TG and LDL in the Tibetan medicine group was significantly decreased(p<0.05 or p<0.01),HDL was significantly increased,the difference was statistically significant(p<0.01);There was no difference in TC(p>0.05).5.Observation of pathological changes of kidney under light microscope and electron microscope: Compared with the control group,the renal tissue of the model group was significantly damaged.Compared with the model group,the Tibetan medicine group significantly improved the renal pathological damage of DN rats.6.Immunohistochemical staining and Western blot was used to detect the changes of inflammatory factors in renal tissues:Compared with the control group,the expression of NF-κB P65、RAGE、TNF-α、IL-6、IL-1β protein in renal tissue of model group was significantly increased(p<0.05);Compared with model group,the expression of NF-κB P65、RAGE、TNF-α、IL-6、IL-1β protein in renal tissue of rats in Tibetan medicine group was significantly decreased(p<0.05).Conclusion:1.Eighteen flavor myrobalan diuretic pills can improve the renal function of DN rats to reduce blood glucose and regulate lipid,improve the pathological damage of DN rats kidney.2.Eighteen flavor myrobalan diuretic pills can can protect kidney tissue by inhibiting the inflammatory pathway of RAGE-NF-κB. |
