Objective: To detect the expression of vascular endothelial growth factor receptor-3(VEGFR-3)and microchromosome maintenance protein 2(MCM2)in esophageal squamous cell carcinoma and normal esophageal tissue,to study the microlymphatic vessel density(MLVD),labeled by monoclonal antibody D2-40 in esophageal squamous cell carcinoma and normal esophageal tissue,and to explore the relationship between microlymphatic vessel density and clinicopathological features and prognosis of esophageal squamous cell carcinoma.To provide meaningful experimental basis for clinical prediction of lymphatic metastasis of esophageal squamous cell carcinoma.Methods: Collected 50 cases of surgical esophageal squamous cell carcinoma tissues and 20 cases of normal esophageal tissue specimens with complete clinical data in 2015 from Suining Central Hospital.Immunohistochemical staining techniques were used to detect the expression of VEGFR-3 and MCM2 at the protein level,and D2-40 was calculated at the same time Labeled microlymphatic vessel density(MLVD),combined with patient clinical data,used x2 test and Fisher exact probability method to explore the correlation between the three and clinicopathological characteristics,and further analyzed the relationship between the three and the esophagus through the Kaplan-Meier method and the R survival package The prognosis of patients with squamous cell carcinoma was correlated,and the survival curve was drawn.Finally,Spearman correlation analysis and log-rank test were used to compare the expression of VEGFR-3 and MCM2 with D2-40 labeled MLVD.Results: 1.The positive expression rate of VEGFR-3 was 58%,which was significantly higher than that in adjacent normal tissues.Its expression was related to lymph node metastasis,but not to age,sex,tumor location,clinical stage,differentiation,smoking history and T stage.The average survival time of 29 patients with esophageal squamous cell carcinoma with high expression of VEGFR-3 was 26.75 months,which was significantly higher than that of patients with low expression of OSCC(39.75months),and there was significant difference in 3-year and 5-year survival rates between the two groups(P < 0.05).In the VEGFR-3 positive expression group,the positive expression rate of MLVD was 93.1%,which was higher than that of the negative expression group(52.4%),and the difference was statistically significant(P < 0.01).There was a positive correlation between VEGFR-3 and MLVD.2.The positive expression rate of MCM2 was 74%,which was significantly higher than that in adjacent normal tissues.Its expression was related to clinical stage,lymph node metastasis and N stage,but not to age,sex,tumor location,differentiation degree and smoking history.The average survival time of 37 patients with esophageal squamous cell carcinoma in MCM2 high expression group was 28.05 months,which was significantly higher than that in low expression group(44.00 months),and there was significant difference in 3-year and 5-year survival rates between the two groups(P < 0.05).In the MCM2 positive expression group,the positive expression rate of MLVD was 78.4%,which was higher than that in the negative expression group(61.5%).There was no significant difference between MCM2 and MLVD.3.The positive rate of MLVD was 76.0%,which was significantly higher than that of normal tissues adjacent to cancer.Its expression was related to lymph node metastasis,but not to age,sex,tumor location,clinical stage,degree of differentiation,smoking history and T stage.The average survival time of 38 patients with MLVD-positive esophageal squamous cell carcinoma was 28.84 months,which was significantly higher than that in the negative group(42.83 months),and there was significant difference in 3-year and 5-year survival rates between the two groups(P < 0.05).Conclusion: This study found that VEGFR-3,MCM2,and D2-40 labeled MLVD in esophageal squamous cell carcinoma were significantly up-regulated compared with normal esophageal tissue adjacent to cancer,and were closely related to tumor lymph node metastasis,and negatively related to survival,and confirmed VEGFR-3 There is a positive correlation with D2-40 labeled MLVD,suggesting that the three play an important role in the development and prognosis of esophageal squamous cell carcinoma.However,this study still has some potential limitations.The limitation of the number of subjects requires multicenter large sample analysis the results are confirmed.Despite the above-mentioned shortcomings,this study provides new ideas for early diagnosis,progress monitoring,prognostic evaluation and finding new therapeutic targets for esophageal squamous cell carcinoma.
