Objective:In this experiment,by studying the expression and correlation of HOXA11,Akt,p-Akt and MMP-2 in pelvic organ prolapse(POP)ligament tissue,explore the role of HOXA11 and PI3K/Akt signaling pathways in POP,and clarify the role of POP The occurrence and development may be related to HOXA11-mediated PI3K/Akt signaling pathway to regulate the extracellular matrix of pelvic floor tissue,which provides a new direction of molecular biology for the clinical prevention and treatment of pelvic floor tissue.Methods:In this study,20 patients in the POP group were selected from the Second Hospital of Shanxi Medical University,who received total laparoscopic hysterectomy +colposacrodesis from October 2019 to December 2020.20 non-POP patients who underwent transabdominal or laparoscopic total hysterectomy in our hospital for benign diseases during the same period were selected as the control group.The selected patients had no history of malignant tumors,no history of endometriosis,no history of pelvic floor prolapse disease and pelvic floor surgery,no history of hormone-related diseases,and 3 history of no hormone medication before surgery.After review and approval by the ethics team,the patient’s sacral ligament tissue was taken during the operation after the patient and family members agreed.The pathological characteristics of the ligament were confirmed by HE and special staining.The activity of HOXA11,Akt,p-Akt and MMP-2in the sacral ligament tissue was detected by immunohistochemistry;Protein expression was detected by WB.Results:1.Masson staining to observe histological characteristics: it can be seen that the collagen fibers are blue-green,and the muscle fibers are red.Microscopic observation of the uterosacral ligament showed that a large number of reticulated cross and regular arranged collagen fibers and regular arranged smooth muscle cells could be seen in the control group,while the collagen fibers in the POP group were significantly reduced with disordered distribution and loose structure.2.Immunohistochemistry:HOXA11,MMP-2,Akt and p-Akt were mainly expressed in the nucleus of the sacral ligament.Compared with the control group,HOXA11 expression in the POP group was significantly weaker,and MMP-2,p-Akt expression,p-Akt / Akt ratio remarkable growth,Akt expression was no different.HOXA11 is negatively correlated with MMP-2 and p-Akt.3.Western-Blot : Compared with the control group,the expression of HOXA11 in the sacral ligament tissue of the POP group was weakened,the expression of MMP-2,p-Akt and the ratio of p-Akt/Akt showed an increasing trend,Akt expression was no different.HOXA11 is negatively correlated with MMP-2 and p-Akt.Conclusion:1.The decrease of HOXA11 expression level is closely related to the occurrence of pelvic organ prolapse.2.Decreased expression of HOXA11 in the human body mediates PI3K/Akt signaling pathway to act on the occurrence of pelvic organ prolapse.3.Phosphorylation of Akt is present in the tissues of POP patients.
