Background The aberrant expression of FRAT1 contributes to poor tumor prognosis in a number of carcinomas.However,its role in glioma is still controversial.ObjectiveTo elucidate the possible relationship between STAT1 and FRAT1,and construe the STAT1 quantity in glioma cells through gene expression profiling.MethodsThe gene expression profiling,KEGG,GO and IPA were used to evaluate the altered expression of genes and proteins in glioma cells post FRAT1 knockdown.Western blot analysis to access the quantity of crucial modules and genes.ResultsIn total,895 down-regulated genes were identified in FRAT1 silenced U251 cells.The most collected procedures as demonstrated by GO and KEGG analysis of 895 unequally status of genes were involved in tumor characteristics.The major collected pathways of these genes in accordance with IPA are considerably magisterial signaling pathways,such as the interferon,and Wnt/β-catenin.And the elevated status of STAT1 in U251 cells was validated.ConclusionThese findings highlighted FRAT1 is the positive regu lator of STAT1,STAT1 can mediate the cancer-promoting effect of FRAT1,FRAT1 through STAT1 to play a cancer-promoting role.
