Orange peel soup was first recorded in Synopsis of Prescriptions of the Golden Chamber and is a famous prescription of traditional Chinese medicine created by Zhang Zhongjing,who is called the medical sage of the Han dynasty.It is mainly composed of two herbs,orange peel and ginger.Orange peel soup is widely used due to its simple composition,convenience and small adverse reaction.The main function of orange peel soup is to stop stagnation and vomiting.At present,the research on orange peel soup mainly focuses on the pharmacokinetics of ginger,orange peel and orange peel soup,along with the clinical effects of anti-vomiting.However,there is few research on the effect of orange peel soup in the intestinal tract,through which the orange peel soup is mainly absorbed.Therefore,this topic is based on the the designed principle of the simulator(SHIME)created by university of Ghent in Belgium to improve the human intestinal microecology system for establishing a gastrointestinal simulation system in vitro suitable for Oriental life habits,with short chain fatty acids as determining index,analyzing the effects of ginger,orange peel,orange peel soup on the digestion in gastrointestinal simulation system in vitro.The 16 Sr RNA high throughput sequencing technique was used to analyze the effect of orange peel soup on intestinal microflora.The purpose of this study was to analyze the effect mechanism of orange peel soup on gastrointestinal tract.Orange peel soup was found to have a positive effect on human health.The main findings are as follows:(1)There was difference between the experimental group and the blank control group.After intervention of orange peel soup,the contents of acetic acid,butyric acid,isobutyric acid,valeric acid and isovaleric acid in the colons were increased while the contents of propanoic acid were decreased,among which the contents of acetic acid,isobutyric acid and butyric acid were significantly increased(P<0.05).After intervention of ginger,the contents of acetic acid,propanoic acid,butyric acid,isobutyric acid,valeric acid and isovaleric acid in the colons were increased,among which the contents of isobutyric acid,butyric acid,iso-valeric acid and valeric acid were significantly increased(P<0.05).After intervention of orange peel,the contents of acetic acid,butyric acid,isobutyric acid,valeric acid and isovaleric acid in the colons were increased,while the contents of propanoic acid were decreased,among which the contents of isobutyric acid,butyric acid,iso-valeric acid and valeric acid were significantly increased(P<0.05).(2)The intestinal flora diversity in ascending colon increased after the intervention of orange peel soup.The relative abundance of Megamonas and Bacteroidetes increased,while the relative abundance of Bifidobacteria,Veillonella,Klebsiella and Shigella escherichia decreased.The intestinal flora diversity in horizontal colon increased after the intervention of orange peel soup.The relative abundance of the Bifidobacteria and Bacteroidetes in the horizontal colon increased,while the relative abundance of Megamonas,Veillonella,klebsiella and Shigella escherichia decreased.The intestinal flora diversity in descending colon increased after the intervention of orange peel soup.The relative abundance of Magamonas and Bifidobacteria in the descending colon increased,while the relative abundance of Bacteroidetes,Veillonella,Klebsiella and Shigella escherichia decreased.In all colons,The relative abundance of Megamonas increased during the washout period.The relative abundance of Bifidobacteria decreased during the washout period.The relative abundance of Veillonella continued to be decreased,and the relative abundance in the washout period decreased by more than 75% compared with the stable period.The relative abundance of Klebsiella decreased by more than 50% in the treatment period compared with the stable period.The relative abundance of Shigella escherichia decreased by more than 70%compared with the stable period.Orange peel soup inhibited the growth of harmful bacteria and regulated the composition of intestinal flora.
