| Background Bone metastasis is one of the common complications of many advanced cancers,of which more than 80% of patients with bone metastasis will develop bone pain,which seriously affects the quality of life of patients.Objective To explore the molecular mechanism of Chinese herbal medicine Huachansu and Kushen on bone cancer pain by the network pharmacological method.Methods Part I The effects of Huachansu and Kushen on pain behavior and bone destruction in bone cancer pain rats were detected.Part II Information on active compounds was obtained from widely cited research and public databases like CNKI,VIP,and TCMSP.The parameters and molecular structures of the active component were obtained from Pub Chem and Chem BL databases,respectively.The molecular structure of the active compounds was imported into the Swiss database to get the predicted targets,and the known therapeutic targets of bone cancer pain were screened by CTD and TTD databases.After name correction of all targets was carried out using the Uniprot database,the intersection of predicted targets and known therapeutic targets of diseases was imported into the String database to screen the core targets of the network.Then,Cytoscape software was used to construct the “drug-target-disease” network for data visualization,and CTD database was used to enrich the biological process of GO-BP and KEGG pathway,to predict the potential mechanism of action of Huachansu and Kushen in the treatment of bone cancer pain.Results Huachansu and Kushen could increase the thermal pain threshold and mechanical pain threshold of the model group and reduce the damage of bone in the rat model of bone cancer pain model group.The 11 active components and 37 core targets of Huachansu were screened out.The GO-BP functional enrichment analysis obtained 807 biological processes(p<0.01);Enrichment analysis of signaling pathways obtained 236 significant correlation pathways(p<0.01).A total of 28 active components and 32 core targets of Kushen were screened.GOBP enrichment analysis obtained 895 biological processes(p<0.01);Signaling pathways were analyzed,and 195 significant correlation pathways(p<0.01)were obtained.Conclusion In this study,the network pharmacological research method was adopted to explore the multi-component,multi-target,and multi-pathway mechanism involved in the treatment of bone cancer pain by traditional Chinese medicine Huachansu and Kushen,providing a basis for further experimental verification and laying a foundation for the clinical research and development of drugs for the treatment of bone cancer pain. |
