Studies On The Chemical Constituents And Quality Control Standard Of Tibetan Drug Herba Arenariae | | Posted on:2022-02-16 | Degree:Master | Type:Thesis | | Country:China | Candidate:D D Song | Full Text:PDF | | GTID:2504306521998049 | Subject:Pharmacy | | Abstract/Summary: | PDF Full Text Request | | Herba Arenariae is the dried roof of perennrial herbage which belong to the Arenariae L.of the family Caryophyllaceae.The Herba Arenariae is a kind of herbal medicine with sweet and cold taste and nourishing yin and blood,and is mainly used in pneumonia and various lung diseases.At present,the basic research on Herba Arenariae is still less,and its quality standard has not been improved,which is not conducive to the development and application of this kind of medicinal materials in China.The aim of this article is to found a group of valid quality standard basis,the following studies were carried out.In order to understand the chemical constituents of Herba Arenariae,in order to provide a basis for the follow-up development of quality standards.In this experiment,the components of Arenaria bryophylla Fernald.were analyzed and identified based on UHPLC-QTOF-MS/MS technology,and a reliable method for compound analysis was established.By obtaining accurate primary and secondary mass spectrometry data,using PeakView 1.2 qualitative software analysis technology,referring to the reported literature,online database and so on,the cracking mode and law of the plant components were summarized.The structures of 60 compounds were inferred in the Arenaria bryophylla Fernald,including 40 flavonoids and 20 other compounds.In this experiment,the chemical constituents of Arenaria bryophylla Fernald were comprehensively analyzed,which provided a basis for the extraction,separation and quality standard study of chemical constituents of Arenaria bryophylla Fernald.Based on the results of LC-MS data analysis of Arenaria bryophylla Fernald.,we continued to separate and identify the chemical components of ethyl acetate extracted from the plant alcohol extract,using a variety of chemical analysis techniques,combined with the corresponding separation methods.20 of them were identified by physical and chemical properties and spectral data.20 compounds were identified by modern chromatographic techniques and a variety of separation methods.They were identified by physicochemical properties and spectral data.Twenty compounds were isolated and identified as tricin(1),askaempferol(2),quercetin(3),isoquercitrin(4),apigein(5),cynaroside(6),β-hydroxypropiovanillone(7),caredhieacid(8),hexadecanoic acid(9),ferulate(10),Vanillate(11),p-hydroxybenzaldehyde(12),formononetin(13),mannitol(14),D-sucrose(15),isovitexin(16),4-hydroxyacetophenone(17),quinic acid(18),caffeic acid(19),hydroquinone(20).Compounds 1-20 are isolated from Arenaria bryophylla Fernald for the first time,compounds 7,11-15,17 are isolated from the Arenaria L.for the first time.Based on the comprehensive study of the chemical constituents of Arenaria bryophylla Fernald and literature investigation,the content of Tricin in Herba Arenariae was assignmented by High Performance Liquid Chromatography.Among them,The method for determination of Tricin is that the separation was performed in a ODS column(Inertsil?ODS-SP,250mm×4.6mm,5μm)with a mobile phase of ACN–0.2%formic acid(35:65).The current speed was 1.0ml/min,and the checking wavelength was at 350nm.The linear range of ellagic acid 0.0400~0.0800μg,the peak area Y and injection capacity X was a fine linear relationship,the regression equation was Y=284876X-18561,with:R~2=0.9999.The content of Tricin in 12 batches of Herba Arenariae from different areas was determined by HPLC.It was stipulated that the content of Herba Arenariae should not be less than 0.12mg/g.The results show that the method is simple,precise and reproducible,and can be used for the quantitative analysis of Herba Arenariae.TLC identification of Tricin is established by using thin layer chromatography,and The moisture,total ash,acid insoluble ash and extract in medicinal materials were determined.The consequence display that the moisture content should be smaller than 14%;the total ash content should be smaller than 10%;and the acid insoluble ash content should be smaller than 5%;the alcohol-soluble extract should be greater than 2.0%.Through the above research,a perfect quality standard of Herba Arenariae was formed,which laid a foundation for the quality evaluation of Herba Arenariae medicinal materials and the utilization of medicinal resources. | | Keywords/Search Tags: | Herba Arenariae, Arenaria kansuensis Maxim, Arenaria bryophylla Fernald, Chemical components, Quality standard | PDF Full Text Request |
