The detection of drug concentration is an important task in clinical and forensic identification,which aims to determine the content of drugs in a pending sample through various detection methods.With the development of society,the requirements for drug systems do not only stop at qualitative testing,but also require drug systems to achieve rapid,stable,and portable quantitative drug testing.The upconversion luminescent material can convert low-energy fluorescence into high-energy fluorescence through photon excitation,and has the advantages of long fluorescence lifetime,narrow emission peak,low toxicity,anti-Stokes frequency shift and anti-biological autofluorescence.Therefore,this article designs and makes a morphine detection system based on the existing upconversion luminescent materials to make it better applicable to the field of morphine detection.The upconversion luminescent system designed in this paper is an optical biosensor that uses upconversion luminescent nanoparticles as a marker and consists of an immunochromatographic test strip and an up-turning luminescence analyzer.The upconversion luminescent analyzer is mainly composed of photoelectric measurement system,control module,algorithm module and mechanical structure.By measuring the intensity and distribution of the upconversion luminescent signal of the upconversion luminescent nanoparticles on the immunochromatographic test strip,the concentration of morphine in the sample to be tested can be determined.This article first introduces the preparation methods of up-conversion luminescence nanoparticles and immunochromatographic test strips,studies the working principles and key technologies of up-conversion luminescence analyzers,designs and manufactures up-conversion luminescence analyzers,and analyzes up-conversion luminescence.The signal collected by the instrument is processed to improve the sensitivity and stability of the system.The up-conversion luminescence system designed in this paper has a better detection limit(0.1ng/ml)and strong stability(the coefficient of variation CV% is maintained below6%).At the same time,because the data collected by the up-conversion luminescence analyzer is interfered by noise,this paper performs binary processing and closed operation on the gray-scaled data,and multiplies the result of the closed operation with the gray-scale data,which is effective Eliminate image noise interference.The adaptive functional boundary algorithm is used to determine the starting point of the fluorescence signal of the control line and the test line,and calculate the ratio of the fluorescence intensity of the control line and the test line to determine the concentration of the target analyte morphine in the sample to be tested.Reduce the error of data processing,and further improve the sensitivity and accuracy of system detection.The main work of the paper is as follows:(1)Research the existing morphine detection methods.(2)The preparation method of upconversion luminescent nanoparticles was studied.Among them,the thermal hydrolysis method is mainly studied.Utilizing ultra-thin m Si O2 to wrap upconversion luminescent nanoparticles(UCNPs)to form a core-shell structure of UCNPs @ m Si O2 has good uniformity,dispersion and stability,while maintaining strong fluorescence emission,improving the sensitivity and accuracy of the test strip.(3)The preparation method of upconversion luminescent material test strips was studied.Among them,the sandwich law and competition law are mainly studied.Aiming at the shortcomings that the morphine molecule is relatively small and the sandwich method cannot make the morphine fully bind to the specific protein carrying the upconversion luminescent material,which makes the detection result inaccurate,this article makes an immunochromatographic test strip based on the competition method.This method can mark the test object as much as possible.Experiments show that the test strip has high stability and reproducibility.(4)The design and manufacture of the upconversion luminescent system structure are studied.The system mainly includes four parts,namely the motion structure,the light source,the main control module,and the image collection module.The system has strong stability and can realize the quantitative detection of morphine.(5)Research the image data processing method.The noise generated by image acquisition has a negative impact on the detection results of the detection system.We solve this problem by adding an algorithm for image data noise reduction,which can make the detection accuracy meet clinical standards.At the same time,smoothing the fitted image data makes the calculation result more accurate.
