Effects Of MiR-1271 On Proliferation And Metastasis In Ovarian Cancer Cells By Targeting Twist1

Ovarian cancer is the fifth leading death in gynecological malignancy,and metastasis is an important cause leading to death.Therefore,it is necessary to explore the pathogenesis of tumor progression,to find out the methods for early diagnosis,and to develop new effective treatment strategies to improve the survival rate and life quality of ovarian cancer patients.Recent studies have shown that the expression of miR-1271 is significantly different in ovarian cancer tissues with normal ovarian tissues,but its potential biological functions and molecular mechanisms in ovarian cancer have not been fully explained.In this study,the expression of miR-1271 in ovarian cancer cells was verified by q RT-PCR.To explore the effects of miR-1271 on proliferation,invasion and migration abilities of ovarian cancer cells,the Cell activity was detected by MTT assay,the cell migration and invasion abilities were measured by Transwell assay,and protein expression was detected by Western Blot.Target of miR-1271 were determined by q RT-PCR,Western Blot and Luciferase reporter assay.The results showed that the expression of miR-1271 in ovarian cancer cells CAOV-3 was down-regulated by comparing with normal ovarian epithelial cells(P <0.05).Twist1 was a target of miR-1271.Over-expression of miR-1271 inhibited the proliferation,migration and invasion of ovarian cancer cells,decreased the expression of cyclin D1,MMP-2 and IL-6,and promoted expression of p21 and E-cadherin(P <0.05),and this effect was restored by overexpression of Twist1.By targeting Twist1,miR-1271 regulates the expression of proliferation and migration-related genes,inhibits the proliferation,migration and invasion of ovarian cancer cells,and can provide a new target and directions for the diagnosis,prevention and treatment of ovarian cancer.
