| At present,the number of deaths from liver cancer each year in the world exceeds half of the total number of liver cancers.At present,surgical resection combined with other treatments is the most important way to treat liver cancer.In this study,hepatoma cell lines Hep3 B cells and Hep G2 cells were cultured in vitro.In this study,we treated these two hepatocarcinoma cell lines with STE.Using MTT,DAPI staining,flow cytometry,laser confocal microscopy,western blotting and other experimental techniques,nude mouse tumor formation experiments were carried out in vivo to enhance the reliability of the experiment,and finally analyze the results in combination with relevant biological statistics.After a series of experimental results analysis,the following conclusions are drawn:1)The morphology of hepatocellular carcinoma cells changes significantly after STE treatment of hepatocellular carcinoma cells.2)Through transcriptome analysis,genes related to apoptosis and autophagy were found,and these genes are related to the death of liver cancer cells.3)The apoptosis-related genes were detected,and it was found that the STE regulated XIAP and further regulated the apoptosis of liver cancer cells.4)STE induces autophagy in liver cancer cells,and after autophagy inhibitors inhibit the autophagy pathway,cell apoptosis is reduced.It shows that autophagy can promote the apoptosis of liver cancer cells.5)STE blocks the cycle of liver cancer cells in G2/M phase.STE inhibits the growth and proliferation of liver cancer cells,and effectively inhibits the cloning ability of liver cancer cells.6)Through the analysis of the experimental results,it is found that in vitro experiments can effectively inhibit the metastasis and invasion of liver cancer cells.7)In vivo experiments have proved to a large extent that STE can inhibit the proliferation of liver cancer cells and promote the apoptosis of liver cancer cells in vivo.In conclusion,STE can down-regulate the expression of XIAP and promote the apoptosis and autophagy of liver cancer cells,eventually leading to cell death.It shows that STE has a therapeutic effect on liver cancer. |
