| Objective: The purpose of this study was to investigate the effects of jiaji electroacupuncture combined with nerve mobilization on the expressions of NF-kB and Mur F1 in gastrocnemius atrophy with sciatic nerve injury in rabbits,so as to find a new way to relieve denervated skeletal muscle atrophy.It provides a theoretical basis for the clinical treatment of denervated muscular atrophy and a new idea and method for the rehabilitation of peripheral nerve injury.Methods: 180 New Zealand large-eared rabbits,half male and half female,were randomly divided into 5 groups: normal control group,model control group,nerve mobilization group,Jiaji electroacupuncture group,Jiaji electroacupuncture combined with nerve mobilization group(referred to as "combination group"),with 36 rabbits in each group.According to 1,2 and 4weeks after surgery,they were divided into 3 subgroups with 12 rats in each subgroup.Using the neural clamps,causing rabbit sciatic nerve in his left leg Sunderland Ⅲ degrees of damage.Normal control group and model control group did not intervene.3 days after surgery,Jiaji electroacupuncture group,nerve mobilization group and combination group were treated with electroacupuncture,nerve mobilization and electroacupuncture combined with nerve mobilization.The behavioral scores of big-eared rabbits were performed by toe reflex and modified Tarlow test at three time points of 1week,2 weeks and 4 weeks,respectively.Then the samples were collected and the cross-sectional area of rabbit gastrocnemius muscle fibers was determined by HE staining.The mRNA expression of NF-kB and Mur F1 was detected by RT-PCR.The protein expressions of NF-kB and Mur F1 were detected by Western Blot.Results:1.Behavioral score of rabbitsAt the time points of 1,2 and 4 weeks,toe reflex and modified Tarlov scores of big-eared rabbits in all groups(except the normal control group)were lower at the first week,and gradually increased with time.At the same time,the behavioral scores of the nerve mobilization group,the Jiaji electroacupuncture group and the combination group were higher than that of the model control group(P<0.05)and lower than that of the normal control group(P<0.05),and the difference was statistically significant.The behavioral scores of the combined group were higher than those of the Jiaji electroacupuncture group and the nerve mobilization group,and the difference was statistically significant(P<0.05).There was no significant difference between Jiaji electroacupuncture group andnerve mobilization group(P<0.05).2.Cross-sectional area of muscle fibersAt the time points of 1,2 and 4 weeks,the reduction degree of cross-sectional area of muscle fibers in all experimental groups(except the normal control group)gradually increased with time,and the atrophy was serious in all experimental groups at the fourth week.At the same time point,nerve mobilization group,Jiaji electroacupuncture group and combination group were lower than normal control group,and higher than model control group(P< 0.05).At the same time point,the combination group was higher than the nerve mobilization group and Jiaji electroacupuncture group(P<0.05);At the same time point,there was no significant difference between the nerve mobilization group and the electroacupuncture group(P >0.05).3.The mRNA and protein expression of NF-kBAt the time point of 1,2 and 4 weeks,the mRNA and protein expression of NF-kB in model control group,nerve mobilization group,Jiaji electroacupuncture group and combination group were all higher than those in normal control group,and reached the peak at 1 week,with statistical significance(P< 0.05).At the same time point,nerve mobilization group,Jiaji electroacupuncture group and combination group were all lower than model control group(P< 0.05).At the same time point,the combination group was lower than the nerve mobilization group and Jianji electroacupuncture group(P< 0.05);At the same time point,there was no statistical significance between electroacupuncture group and nerve mobilization group(P >0.05).4.The mRNA and protein expression of Murf1At the time point of 1,2 and 4 weeks,the mRNA and protein expression of Murf1 in model control group,nerve mobilization group,electroacupuncture group and combination group were all higher than those in normal control group,and reached the peak at 1 week,with statistical significance(P< 0.05).In the 1 week group,there was no significant difference among the nerve mobilization group,the Jiaji electroacupuncture group,the combination group and the model control group(P> 0.05).In the 2and 4 week groups,the nerve mobilization group,the Jianji electroacupuncture group and the combination group were lower than the model control group(P<0.05),and the combination group was lower than the nerve mobilization group and the Jianji electroacupuncture group(P<0.05).There was no statistical significance between the nerve mobilization group and the Jianji electroacupuncture group(P> 0.05).Conclusion1.Both Jiaji electroacupuncture and nerve mobilization can relieve gastrocnemius atrophy with sciatic nerve injury in rabbits,and the combined effect of both is better.2.Jiaji electroacupuncture combined with nerve mobilization relieve gastrocnemius atrophy with sciatic nerve injury in rabbits,and the mechanism may be related to the inhibition of NF-kB and Murf1 expression in the NF-kB/Murf1 pathway. |
