Objective This article aims to investigate the expression of Notch1 and Hes1 in Diffuse large B cell lymphoma,and its correlation with clinical characteristics of DLBCL patients.Methods Clinical data of 54 newly diagnosed DLBCL patients in Hebei People’s Hospital from June 2017 to September 2020 were collected.The expression of Notch1 and Hes1 in DLBCL by immunohistochemistry.SPSS 26.0 statistical software was used to analyze the relationship between the expression of Notch1 and Hes1 protein and the clinical characteristics of DLBCL(age,B symptoms,Ann-Arbor stage,etc.).The difference was statistically significant in P<0.05.Results 1.In this study,54 patients with DLBCL were collected.According to gender : 35 cases(64.8%)in male and 19 cases(35.2%)in female;and the ratio of male to female was about 1.8:1;18 cases(33.3%)in ≤60 years and 36 cases(66.7%)in > 60 years;The age of onset ranged from 10 to 88 years old;The median age of onset was 68 years old;34cases(63.0%)were without B symptoms,and 20 cases(37.0%)were with B symptoms.According to Ann-Arbor stage : 25 cases(46.3%)were in Ann-Arbor Ⅰ/Ⅱ stage,29 case(53.7%)were in Ann-Arbor Ⅲ/Ⅳ stage;In eastern cooperative oncology group score,44 cases(81.5%)scored < 2and 10 case(18.5%)score ≥ 2;According to the International Prognostic Index(IPI),35 cases(64.8%)scored 0~2,and 19 cases(35.8%)scored 3~5;According to Hans subtype: 34 cases(64.2%)were of germinal center B-cell-like GCB subtype and 34(63.0%)were non-germinal center B-cell-like subtype;Lymphocyte count was normal in 34 cases(63.0%),decreased in 19 patients(35.2%)and elevated in 1 patients(1.8%);the lactate dehydrogenase level was normal in 25 cases(46.3%)and was elevated in 28 cases(51.9%)and decreased in 1 patients(1.8%);According to the level of Ki-67,there were 8 patients(14.8%)with Ki-67 < 60%,and46 patients(85.2%)with Ki-67 ≥60%.2.In DLBCL group,the positive rate of Notch1 was 51.9%(28/54),the negative rate was 48.1%(26/54);In the control group,the positive rate of Notch1 was 15.0%(3/20)while the negative rate of Notch1 was 85.0%(17/20),The positive rate of Notch1 in DCBCL group was significantly higher than that in control group,and the difference was statistically significant(P<0.05);The positive rate of Hes1 was 61.1%(33/54)while the negative rate was 38.9%(21/54).In the control group,the positive rate of Hes1 was 30.0%(6/20)while the negative rate of Hes1 was 70.0%(14/20).The positive rate of Hes1 in DCBCL group was significantly higher than that in control group,and the difference was statistically significant(P<0.05).3.The positive rate of Notch1 was correlated with B symptoms,Ann-Arbor stage,lymphocyte count and lactate dehydrogenase(P<0.05).Notch1 was highly expressed in B symptom group,Ann-Arbor stage Ⅲ / Ⅳ,decreased lymphocyte count group and elevated LDH group.The expression of Notch1 protein was not correlated with gender,age,ECOG score,IPI score,Hans type and Ki-67 index(P>0.05).4.The positive rate of Hes1 was correlated with B symptoms;The positive rate of Hes1 in B symptoms group was higher than that without B symptoms;The expression of Hes1 protein was not correlated with gender,age,Ann Arbor stage,ECOG score,IPI score,Hans type,lymphocyte count,lactate dehydrogenase and Ki-67 index(P > 0.05).5.Notch1 and Hes1 were all positive in 21 cases(38.9%)and all negative in 14 cases(25.9%)of DLBCL.According to Spearman rank correlation analysis,Notch1 was correlated with Hes1(r =0.296,P<0.05).Conclusions 1.The expressions of Notch1 and Hes1 in DCBCL patients were significantly higher than those in the control group,and was related to B symptoms and Ann-Arbor stage.These results suggest that they may play an important role in the occurrence and development of DCBCL.2.There was a positive correlation between the expression of Notch1 and Hes1 in DCBCL patients,suggesting that Notch1 and Hes1co-expressed play a synergistic role in promoting tumor.
