Esophageal carcinoma is highly occurred in china and is a worldwide disease that threaten to public health with high mortality.Towards esophageal cancer,in clinic four therapy methods are adopted for now.Through meta-analysis,we conclude that immunological therapy combining with chemicals perform the best efficiency to treat late-stage patients.For immunological therapy,in this article we mainly discuss about principles and fundamental procedures to conduct with CAR-T.After construct expression plasmid,transfect into Tcells,co-culture with cancer cells,utilizing confocal microscope,flow cytometry and MTT test,efficiency of chimeric T cells is researched.In addition,the most significant part of experiment design is to confirm with appropriate antigens.Based on GEO database,along with searching of reports and articles,we screen 4 promising antigens which can be immunological targets.5 genes are related with immunological inhibition.Through survival analysis,methylation analysis and expression information of micro RNA,mechanisms of esophageal carcinoma are discussed in this article.At the last of the article,we introduce about Chinese traditional diagnosis method,namely tongue diagnosis,which can be adopted to assist with earlystage esophageal cancer screening.Molecular mechanisms are mentioned as well.main work in this article include:(1)Meta-analysis to evaluate efficiency of four invention methods Utilizing software to conduct with meat-analysis towards four invention methods in clinic to treat esophageal cancer.The result indicates that little heterogeneity exists among several methods.Among them,immunological therapy combining with chemicals perform best to relieve patients’ illness.(2)Cell test to discuss about function of chimeric T cells After design experiments,construct expression plasmid,transfect into T cells and co-culture with cancer cells,function of chimeric T cells is discussed.(3)Screen promising antigen targets based on GEO database Based on GEO database,four promising antigens are concluded for immunological therapy.structure and function of relative proteins are discussed through analysis with websites and databases.(4)Researches on mechanisms of esophageal carcinoma based on GEO databaseIn addition,we also analyze with down-regulated genes,survival curves,methylation information and micro RNA expression about esophageal cancer.Pathways and signaling are discussed according to specific genes.Through discussion,mechanisms of esophageal carcinoma development are researched.(5)Tongue diagnosis to screen early-stage esophageal cancer As known that abnormal expressed proteins can be viewed as biomarkers for diagnose.In this article,we introduce a new method to screen early-stage esophageal carcinoma.Through data analysis,the result demonstrate that tongue diagnose is an effective method to detect precancerosis esophageal cancer.Molecular mechanisms are discussed in this article to reveal about reasons.To sum up,methods to treat esophageal cancer,immunological therapy method,antigen screening,molecular mechanisms and tongue diagnose are discussed in this article to reveal fundamental principles and mechanisms of esophageal carcinoma.
