Objectives:ABCA1 is a member of the ABC transporter family,which plays a major role in the formation of high-density lipoprotein and cholesterol reverse transport.We aim to study the 69C/T and R219 K single nucleotide polymorphisms of the ABCA1 gene on blood lipids and susceptibility to coronary heart disease.,The influence of the extent of coronary artery disease and the degree of stenosis.Methods:A total of 261 subjects were recruited in this experiment,and they were divided into 127 persons in the coronary heart disease group and 134 persons in the control group.The fasting venous blood of the subjects was collected in the morning to determine the blood lipid level.Using polymerase chain reaction-restriction fragment length polymorphism(PCR-RELP)method,DNA extraction,primer design,polymerase chain reaction(PCR)amplification,DNA purification,Enzyme digestion,agarose gel electrophoresis and other processes,obtained data to analyze the possible associations between the ABCA1 gene 69C/T and R219 K polymorphic variants and blood lipids,coronary heart disease susceptibility,and the extent of coronary artery disease and stenosis.Results:1.The gene distribution frequency of 69C/T polymorphism in the coronary heart disease group was TT(32.3%),CT(47.2%),CC(20.5%),T(55.9%),C(44.1%),while 69C/T polymorphism in the control group The gene distribution frequency of T polymorphism is TT(16.4%),CT(39.6%),CC(44.0%),T(36.2%),C(63.8%),TT genotype and T allele are in the two groups There is a significant difference in the distribution between the coronary heart disease group is significantly higher than the control group,the difference is statistically significant(P <0.05).The levels of blood lipids(LDL-C,HDL-C,TG,TC)of each genotype of 69C/T were compared,and the difference was not statistically significant.In patients with coronary heart disease,the influence of 69C/T on the extent and stenosis of coronary artery disease was further analyzed.The results showed that the distribution frequency of TT genotype and T allele in double-vessel disease and multivessel disease was higher than for single-vessel disease,the difference was statistically significant(P<0.05).The frequency of distribution of moderate and severe coronary lesions was higher than that of mild disease,and the difference was statistically significant(P <0.05).2.The gene distribution frequency of R219 K polymorphism in patients with coronary heart disease was RR(37.0%),RK(48.8%),KK(14.2%),R(61.4%),K(38.6%),while the control group was R219 K polymorphism The gene distribution frequencies of KK are RR(26.10%),RK(40.3%),KK(33.6%),R(46.3%),K(53.7%).The distribution of KK genotype and K alleles between the two groups is Significant difference,the coronary heart disease group was significantly lower than the control group,the difference was statistically significant(P<0.05).The levels of blood lipids(LDL-C,HDL-C,TG,TC)of each genotype of R219 K were compared,and the level of HDL-CC(1.35±0.48)of R219 K KK type was higher than that of RR type(1.03±0.30)and RK type.(1.13±0.32),KK type TG(1.41±0.72)level was lower than RR type(2.01±1.29)and RK type(1.57±0.77),the difference was statistically significant(P<0.05).In the coronary heart disease group,the effect of R219 K on the extent of coronary artery disease and the degree of stenosis was further analyzed,and the difference was not statistically significant(P>0.05).3.Logistic regression analysis of related influencing factors of coronary heart disease showed that the incidence of coronary heart disease in TT gene carriers was 2.3 times that of non-carriers(OR=2.341,95% CI 1.296-4.226,P<0.05);and KK gene carriers The incidence of coronary heart disease in patients was lower than that of non-carriers(OR=-0.689,95%CI was 0.561-0.845,P<0.05);the rest of the analysis of diabetes,genetic family history and smoking history showed that these risk factors can increase coronary The incidence of heart disease(P<0.05).Conclusions:ABCA1 gene 69C/T polymorphism TT genotype and T allele can increase the susceptibility of coronary heart disease,and can aggravate the extent of coronary artery disease and stenosis,but it has nothing to do with blood lipid level;R219K polymorphism KK genotype and K allele can reduce the susceptibility of coronary heart disease,and is related to the level of high-density lipoprotein,but has nothing to do with the extent of coronary artery disease and the degree of stenosis.
