Expression Of PDCD11 In Colorectal Cancer And Its Clinical Significance

Abstract/Summary:
Objective:The aim of this study is to detect the expression of PDCD11 in colorectal cancer(CRC)and to explore whether there is a correlation between its expression level and clinicopathological features.Methods: Samples of colorectal cancer and corresponding paracancerous tissues were collected from 50 patients admitted to the affiliated Hospital of Inner Mongolia Medical University from September 2019 to October 2020.IHC-P was used to evaluate the expression of PDCD11 in CRC tissues and corresponding paracancerous tissues,and to explore the correlation between the expression level of PDCD11 and the characteristics of clinical factors.After that,15 pairs of sample tissues were randomly selected,and the relative expression of PDCD11 mRNA and protein in CRC tissues and corresponding paracancerous tissues were quantitatively detected by q RT-PCR and Western blot.Graph Pad Prism8.0 and SPSS26.0 were used to analyze and process the original experimental data.Results:1.The results of IHC-P showed that among the 50 paired samples,33 cases of CRC tissues were positive for PDCD11,with a positive rate of 66%,and 13 cases of paracancerous tissues were positive for PDCD11,with a positive rate of 26%.In the high power visual field(400 ×),PDCD11 protein was located in the nucleus and distributed in brown granules.2.Through chi-square test and Fisher’s exact test,it was found that there was no significant correlation between the expression level of PDCD11 and sex(P=0.196),age(P=0.276),tumor location(P=0.584),tumor diameter(P=0.480),differentiation(P=0.878),but it was significantly correlated with TNM stage(P=0.033)and lymph node metastasis(P=0.012).3.q RT-PCR results showed that the relative expression of PDCD11 mRNA in CRC tissues was 2.38 ±0.78,which was significantly higher than that in paracancerous tissues(1.55 ±0.65).There was a significant difference in the expression between the two groups(P = 0.004 <0.05).4.Western blot results showed that the expression of PDCD11 protein in CRC tissues was 1.50 ±0.48,which was significantly higher than that in paracancerous tissues(1.14 ±0.38).There was significant difference in expression between the two groups(P = 0.03 < 0.05).Conclusion:The expression of PDCD11 in CRC is higher than that in adjacent tissues,and the expression level of PDCD11 is related to TNM stage and lymph node metastasis.PDCD11 may be a cancer-promoting factor of colorectal cancer and is expected to become a biomarker for the diagnosis and prognosis of colorectal cancer patients in the future.
