Expression And Clinical Significance Of LncRNA TOB1-AS1 In Ovarian Cancer Tissues

Objective: To detect the expression of long non-coding RNA(lnc RNA)transducer of Erb B2,1-antisense 1(TOB1-AS1)in epithelial ovarian cancer(EOC)tissues and normal ovarian tissues.By analyzing the relationships between TOB1-AS1 expression level and the clinic-pathologic features of EOC patients,to further explore the impact of TOB1-AS1 on the clinical prognosis of EOC patients.Methods: From July 2017 to January 2018,the 67 cancer tissues from EOC patients,who were treated at the the Department of Gynecology of the Fourth Hospital of Hebei Medical University were collected.The followed-up observation has been kept until January 31,2021.During the same period,30 normal ovarian tissues of patients with benign gynecological diseases were collected as the control group.The EOC dataset in TCGA(The Cancer Genome Atlas)database was analyzed by statistical test.And the quantitative real-time polymerase chain reaction(q RT-PCR)method was used to detect the expression level of TOB1-AS1 in 67 cases of EOC tissues and 30 cases of normal ovarian tissues.The relationships between TOB1-AS1 expression in EOC tissues and clinic-pathologic features,such as the age,Federation International of Gynecology and Obstetrics(FIGO)stage,tumor differentiation degree,lymph node metastasis,and peritoneal metastasis of patients were analyzed by statistical test.Kaplan-Meier and statistics were used to analyze the relationships between TOB1-AS1 expression level and progression-free survival(PFS)and overall survival(OS)of EOC patients.Results:1.The disparity expression of TOB1-AS1 in EOC tissues and normal ovarian tissues.The analysis of the EOC dataset in TCGA database showed that the expression of TOB1-AS1 in EOC patients tissues was significantly higher than that in normal ovarian tissues(P<0.01).And by q RT-PCR,TOB1-AS1 is highly expressed in EOC tissues compared with normal ovarian tissues(P<0.01).2.The relationships between the expression level of TOB1-AS1 and clinic-pathologic features of EOC patients.Statistical analysis of the relationships between TOB1-AS1 expression in EOC tissues and clinic-pathologic features of patients showed that the expression of TOB1-AS1 was significantly higher in EOC patients with FIGO stage III+IV,poorly differentiated tumor,positive lymph node metastasis,and positive peritoneal metastasis than that of EOC patients with FIGO stage Ⅰ+Ⅱ,middle and high differentiation,negative lymph node metastasis,and negative peritoneal metastasis(P<0.05).However,there was no significant difference between the expression of TOB1-AS1 and the age of patients(P>0.05).3.The survival analysis between the expression level of TOB1-AS1 and the prognosis of EOC patients.According to the expression level of TOB1-AS1,67 EOC patients were divided into high TOB1-AS1 expression group and low expression group.The results of survival analysis showed that the median PFS of the high TOB1-AS1 expression group was significantly lower than that of the low TOB1-AS1 expression group(21 months vs not reached,Log-rank test:P=0.001).By the end of the followed-up,both groups did not reach the median OS,while the Log-rank test indicated that there was a statistical difference in overall survival between the two groups(P=0.025).Furthermore,the 36 th month was used as the cut-off point to compare the differences in prognosis between the two groups in terms of survival.The results showed that the 3-year PFS rate of the high TOB1-AS1 expression group was significantly lower than that of the low TOB1-AS1 expression group(19% vs52%,P=0.005).The 3-year OS rate of the high TOB1-AS1 expression group was also significantly lower than that of the low TOB1-AS1 expression group(50% vs 80%,P=0.015).Conclusions:1.TOB1-AS1 is highly expressed in EOC tissues.2.The expression of TOB1-AS1 in EOC tissues was correlated with the FIGO stage,tumor differentiation,lymph node metastasis and peritoneal metastasis,while there was no obvious correlation with the age of patients.3.The high expression of TOB1-AS1 was associated with poor prognosis of EOC patients.
