Objective: To study the expression of sex determining region Y-box protein 5(SOX5)in oral squamous cell carcinoma(OSCC)tissues and to study its role in the invasion and metastasis of OSCC cell line CAL27 in vitro,with a view to explore the new target of OSCC provides a theoretical basis for the diagnosis and treatment of OSCC.Methods: The Envision two-step immunohistochemical method was used to detect the expression of SOX5 in OSCC tissues and adjacent non-tumor tissues,and the collected clinical pathological data of patients were used to analyze the relationship between SOX5 and the expression level of SOX5 in OSCC tissues.Relationship.Cultivate human normal oral mucosal cell lines HOK,OSCC cell lines CAL27,SCC9,SCC15,SCC25,and clarify the expression of SOX5 in the above five cell lines by Western blot,and select CAL27 as the cells for later research based on the experimental results.Divide CAL27 into Si-NC group and Si-SOX5 group according to the transfection situation,adopt q RT-PCR and Western Blot method to clarify the interference efficiency,adopt plate cloning,scratch and Transwell experiment to detect the effect of down-regulation of SOX5 expression on cell proliferation and migration and invasion ability.Results:1.The expression of SOX5 in OSCC tissues was significantly higher than that in adjacent tissues(P<0.01),and was related to lymph node metastasis(P<0.05),and had nothing to do with gender,age,depth of tumor invasion,TNM stage,and tumor differentiation(P>0.05).2.Western blot showed that compared with HOK,SOX5 was highly expressed in OSCC cell lines CAL27,SCC9,SCC15,and SCC25.3.After Si RNA interference treatment,the m RNA and protein expression of CAL27 Si-SOX5 group was significantly lower than that of Si-NC group.4.In the plate clone formation experiment,compared with the Si-NC group,the number of CAL27 cell colonies in the Si-SOX5 group was significantly delayed and decreased(P<0.01).5.The results of the scratch test showed that at 24 h the Si-SOX5 group had a slower scratch healing rate than the Si-NC group,that is,the migration ability was inhibited(P<0.05),and it is more obvious inhibited that the Si-SOX5 group had a lower migration ability than the Si-NC group at 48h(P<0.01).6.Transwell method migration experiment also showed that the migration of CAL27 in SiSOX5 group was significantly inhibited(P<0.01),and invasion experiment showed that the invasion ability of Si-SOX5 group cells was significantly lower than that of the Si-NC group(P<0.01).Conclusion: SOX5 is highly expressed in OSCC tissues and OSCC cell lines CAL27、SCC9、SCC15 and SCC25.The expression level of SOX5 is correlated with lymph node metastasis of OSCC.The results of measuring the interference efficiency at the m RNA and protein levels show that interfering with SOX5 can significantly inhibit the expression of SOX5 in CAL27 and interfering with SOX5 can inhibit the proliferation,cell migration and invasion of CAL27 cells.
